Organs-on-chips: a decade of innovation

Carly Strelez et al.

Summary: Recreating "living organs" using groundbreaking organ-on-a-chip (OOC) technologies is revolutionizing drug discovery. Studies by Huh et al. and Ronaldson-Bouchard et al. highlight the advancements made over the past decade, from single organ functionality to interconnected organs, allowing for the examination of drug toxicities and disease pathogenesis in reconstituted tissues.

Design and engineering of organ-on-a-chip

Sujin Cho et al.

Summary: Organ-on-a-chip (OOC) is a new interdisciplinary technology that recreates the structure, function, and physiology of human tissues for drug screening purposes. Significant progress has been made in the past decade in replicating tissue and organ functions on chips through advances in biomaterials, stem cell engineering, microengineering, and microfluidic technologies. The critical factors for reconstituting tissue function and microenvironment on chips include structural and engineering constituents, as well as biological components.

Polydopamine and gelatin coating for rapid endothelialization of vascular scaffolds

Kamil Kopec et al.

Summary: Rapid endothelialization is crucial for overcoming the limitations of small-diameter vascular grafts. In this study, biomimetic coatings were developed using medical-grade polyurethane nanofibrous mats and tubular scaffolds. The coatings, consisting of polydopamine and gelatin, significantly enhanced endothelialization on both flat surfaces and small-diameter tubular scaffolds, as well as commercial vascular prostheses.

A human kidney and liver organoid-based multi-organ-on-a-chip model to study the therapeutic effects and biodistribution of mesenchymal stromal cell-derived extracellular vesicles

Vivian V. T. Nguyen et al.

Summary: This study used an in vitro multi-organ-on-a-chip model to investigate the effects of MSC-derived sEVs on kidney and liver organoids. The results showed that sEVs stimulated recovery of injured renal epithelium and accumulated in both injured kidney cells and intact liver organoids. This research method provides insights into the mechanism of action and potential side effects of sEVs.

Summary: The Nephroscreen platform, using a combination of cell viability, LDH, and miRNA release, proved to be a robust and predictive method for detecting drug-induced kidney injury. It is compatible with automated pipetting, suitable for long-term experiments, and offers a valuable tool for replacing animal testing and supporting the 3Rs principles.

Long-Lived Human Lymphatic Endothelial Cells to Study Lymphatic Biology and Lymphatic Vessel/Tumor Coculture in a 3D Microfluidic Model

Nicola Frenkel et al.

Summary: The lymphatic system is vital for maintaining tissue fluid balance and transporting antigens and immune cells to lymph nodes. A microfluidic lymphatic vessel model was developed to investigate lymphangiogenesis and its interaction with colon cancer organoids using immortalized lymphatic endothelial cells. The model provides a stable, standardized platform for studying cancer cell-induced lymphangiogenesis and cancer cell motility.
