Grip strength is inversely associated with DNA methylation age acceleration

Mark D. Peterson et al.

Summary: This study aims to investigate the association between grip strength and DNAm age acceleration. The results show a strong and independent cross-sectional and longitudinal association between grip strength and DNAm age acceleration. Men and women with lower grip strength and loss of strength over time may experience accelerated aging.

Plasma metabolomic signatures of dual decline in memory and gait in older adults

Qu Tian et al.

Summary: Older adults experiencing decline in both memory and gait have a higher risk of dementia compared to those with decline in only one of these factors, and this study aimed to explore the underlying mechanisms. By analyzing plasma metabolite biomarkers, the researchers found that specific metabolic pathways, including lysophosphatidylcholines, ceramides, and amino acids, were significantly altered in older adults with dual decline. These alterations in metabolites may indicate mitochondrial dysfunction, compromised immunity, and increased burden of cardiovascular and kidney pathology.

Body fat and components of sarcopenia relate to inflammation, brain volume, and neurometabolism in older adults

Wouter A. J. Vints et al.

Summary: Obesity and sarcopenia are associated with cognitive impairments in older adults. Blood biomarkers may play a role in mediating this relationship by influencing neurometabolism and brain structure. This study used a multimodal approach to comprehensively assess the mechanism of body-brain crosstalk.

Cerebrospinal fluid neurofilament light chain is a marker of aging and white matter damage

Karin L. Meeker et al.

Summary: CSF NfL is a non-specific marker of aging and white matter integrity, and is more strongly associated with total white matter hyperintensity volume than with gray or white matter volume. Age and white matter hyperintensity volume are consistently more important predictors of CSF NfL than AD markers.

Plasma neurofilament light levels associate with muscle mass and strength in middle-aged and older adults: findings from GenoFit

Jedd Pratt et al.

Summary: This study examined the association between plasma NfL concentration and sarcopenic domains. The results showed that NfL levels were associated with grip strength and SMI and were significantly elevated in sarcopenic individuals. This suggests that NfL levels could be a potential biomarker for sarcopenia.

Longitudinal associations of absolute versus relative moderate-to-vigorous physical activity with brain microstructural decline in aging

Qu Tian et al.

Summary: Higher levels of moderate-to-vigorous physical activity may help preserve brain structural integrity, particularly in selected temporal areas. The relationship between physical activity and brain changes seems to depend on the relative intensity of the activity rather than the absolute amount.

Associations between grip strength, brain structure, and mental health in > 40,000 participants from the UK Biobank

Rongtao Jiang et al.

Summary: This study used data from over 40,000 participants in the UK Biobank to explore the relationship between grip strength, mental health, and brain structure. The results showed that stronger grip strength was associated with better cognitive functioning, higher life satisfaction, and reduced symptoms of depression and anxiety. Grip strength was also related to the volume of grey matter in specific regions of the brain, which in turn correlated with better mental health.

An examination of the relationship among plasma brain derived neurotropic factor, peripheral vascular function, and body composition with cognition in midlife African Americans/Black individuals

Miranda K. Traylor et al.

Summary: Despite higher risk factors for neurodegeneration, African American/Black individuals have been excluded from important neuroscience research. This study found a significant correlation between plasma BDNF levels and complex attention and processing speed, but no significant relationship between vascular measures and cognitive domains or BDNF values. The results suggest that interventions targeting increased BDNF could potentially improve cognition, and interventions promoting skeletal muscle size and function may be beneficial for cognitive health in the African American/Black population.

Neurofilament light chain concentration in an aging population

Aurelie Ladang et al.

Summary: In this study, we assessed the concentration of neurofilament light chain (NF-L) in the serum of 409 community-dwelling adults aged over 65 years. We found that NF-L concentration was associated with kidney function, age, and body mass index (BMI). Subjects with self-reported neurological disorders had higher NF-L concentration. Furthermore, NF-L concentration was also associated with cognitive impairment but not with depression. It is important to interpret NF-L measurements with caution, especially in patients with renal failure.

Summary: The study found that neurofilament light chain protein (NfL) is associated with cognitive function in Parkinson's disease (PD) patients and can predict future cognitive decline. Plasma NfL was identified as a useful prognostic biomarker for PD, predicting the likelihood of PD patients converting to mild cognitive impairment or dementia.

Investigating Associations Between Inflammatory Biomarkers, Gray Matter, Neurofilament Light and Cognitive Performance in Healthy Older Adults

Hollis C. Karoly et al.

Summary: The study explored associations between age, inflammatory markers, neurofilament levels, brain volume, and cognitive performance in healthy older adults. Results indicated that multiple gray matter regions in the brain link age and cognitive performance, suggesting that neurofilament levels alone may not be sufficient markers of brain changes related to aging, inflammation, and cognitive performance.
