rxCOV is a quantitative metric for assessing immunoassay analyte fidelity

Rhonda. M. M. Brand et al.

Summary: Immunoassay based bioanalytical measurements are widely used in biomedical research and clinical applications. However, there is currently no objective metric to assess the accuracy of these measurements. This study introduces a fidelity metric called ratio of cross-coefficient-of-variation (rxCOV) to objectively evaluate the measurement quality of immunoassay analytes in comparing different sample groups or experimental conditions. The feasibility and applicability of rxCOV are demonstrated using simulated and experimental data, showing its independence from statistical significance and ability to identify meaningful differences. Its importance in increasing signal to noise ratio and its potential applications are also discussed. The adoption of rxCOV as a simple and easy-to-use fidelity metric for robust and reproducible biomedical research is anticipated.

The reactome pathway knowledgebase 2022

Marc Gillespie et al.

Summary: The Reactome Knowledgebase, a core resource of Elixir, provides manually curated molecular details of human biological processes, serving as a digital archive and tool for discovering functional relationships. Recent work has expanded annotations of signaling processes, drugs targeting them, infections by coronaviruses, and better analysis tools for high-throughput data and placement of unstudied proteins.

The Different Immune Profiles of Normal Colonic Mucosa in Cancer-Free Lynch Syndrome Carriers and Lynch Syndrome Colorectal Cancer Patients

Lena Bohaumilitzky et al.

Summary: The immune infiltrate in tumor-distant normal colorectal mucosa from individuals with Lynch syndrome (LS) was assessed. It was found that LS carriers show elevated mucosal T-cell infiltration even in the absence of cancer, and this immune profile may impact tumor risk in LS carriers.

Immunology of Lynch Syndrome

Danielle M. Pastor et al.

Summary: The review provides a comprehensive assessment of the immunologic aspects of Lynch syndrome pathogenesis, focusing on the immunogenic properties of Lynch syndrome polyps and associated cancers, as well as potential immune interventions. The rich immune microenvironment of Lynch syndrome lesions offers opportunities for immune-mediating agents to enhance host immune responses and reduce immunosuppressive entities, which can be utilized in prevention and treatment trials for patients with Lynch syndrome polyps and associated cancers.
