FCN3 inhibits the progression of hepatocellular carcinoma by suppressing SBDS-mediated blockade of the p53 pathway

Dong Ma et al.

Summary: Hepatocellular carcinoma (HCC) is the third-leading cause of cancer deaths globally. Downregulation of Ficolin-3 (FCN3) protein in HCC tissues is closely related to poor prognosis. Overexpression of FCN3 induces apoptosis and inhibits cell proliferation via the p53 signaling pathway. FCN3 modulates the nuclear translocation of eukaryotic initiation factor 6 (EIF6) by binding ribosome maturation factor (SBDS) to induce ribosomal stress and activation of the p53 pathway. Y-Box Binding Protein 1 (YBX1) is involved in the transcription and translation level regulation of FCN3 to SBDS. A negative feedback loop involving p53, YBX1, and SBDS is identified downstream of FCN3.

Comprehensive analysis identifies CLEC1B as a potential prognostic biomarker in hepatocellular carcinoma

Qiangan Jing et al.

Summary: CLEC1B, encoding the CLEC-2 protein, is involved in platelet activation, angiogenesis, and immune and inflammatory responses. This study found that CLEC1B is downregulated in hepatocellular carcinoma (HCC) and has a good clinical prognostic value. CLEC1B expression is closely associated with immune cell infiltration and correlates with a variety of immunomodulators in the HCC tumor microenvironment. Furthermore, CLEC1B and its related genes or interacting proteins are implicated in multiple immune-related processes and signaling pathways. Overexpression of CLEC1B significantly influences the treatment effects of sorafenib on HCC cells.

Widespread genomic/molecular alterations of DNA helicases and their clinical/therapeutic implications across human cancer

Xin Qin et al.

Summary: DNA helicases play a crucial role in maintaining genomic stability, and mutations in these genes can lead to genomic instability and cancer susceptibility. Interestingly, overexpression of DNA helicases is observed in certain cancers. In this study, genomic and molecular alterations in 12 important DNA helicases were analyzed across various types of cancers. The study revealed significant expression heterogeneity among these helicases, and tumors were classified into subtypes based on their DNA helicase score. The high DNA helicase score subtypes were associated with stemness, proliferation, oncogenic signaling, longer telomeres, more mutations, copy number alterations, and shorter survival. Furthermore, high DNA helicase scores were linked to sensitivity to chemo- and radio-therapies, as well as defects in DNA repair pathways like homologous recombination deficiency. Importantly, a potential DNA helicase inhibitor, Danusertib, was identified in this study. These findings highlight the dysregulation of DNA helicases in cancer and their potential as therapeutic targets.

Summary: This study investigated the effects of low concentrations of cadmium (Cd) on liver cancer cell proliferation, steatosis, and fibrogenic/oncogenic signaling. Cd exposure increased reactive oxygen species production, enhanced cell proliferation, and activated Notch and AKT/mTOR pathways in liver cancer cells. Amsacrine downregulates BCL2L1 expression and triggers apoptosis in human chronic myeloid leukemia cells through the SIDT2/NOX4/ERK/ HuR pathway

Jing-Ting Chiou et al.

Summary: Accumulating evidence suggests that the anticancer activity of acridine derivatives in leukemia cells is achieved by regulating the expression of BCL2 family proteins, including BCL2L1. Amsacrine inhibits the expression of BCL2L1 in K562 cells, leading to apoptosis. This downregulation is mediated by the SIDT2/NOX4/ERK pathway, which results in the downregulation of HuR and subsequent degradation of BCL2L1 mRNA. This pathway is also observed in other leukemia cell lines.

FCN3 inhibits the progression of hepatocellular carcinoma by suppressing SBDS-mediated blockade of the p53 pathway

Summary: Hepatocellular carcinoma (HCC) is a major cause of cancer-related deaths worldwide, and current treatment strategies have limited effectiveness. This study identified Ficolin-3 (FCN3) as a potential prognostic factor for HCC, with decreased expression associated with poor prognosis. Overexpression of FCN3 was found to induce apoptosis and inhibit cell proliferation through the p53 signaling pathway. Furthermore, FCN3 was shown to modulate nuclear translocation of EIF6 by binding to SBDS, leading to ribosomal stress and activation of the p53 pathway. Y-Box Binding Protein 1 (YBX1) was also found to regulate FCN3 and SBDS at the transcription and translation level. Additionally, a negative feedback loop involving p53, YBX1, and SBDS was identified downstream of FCN3.
