One-Pot Facile Encapsulation of Dimethyloxallyl Glycine by Nanoscale Zeolitic Imidazolate Frameworks-8 for Enhancing Vascularized Bone Regeneration

Xin Zhang et al.

Summary: In this study, dimethyloxallyl glycine (DMOG) was loaded onto nanoscale zeolitic imidazolate frameworks-8 (ZIF-8) to create a drug-loading system that promotes osteogenesis-angiogenesis coupling. The results showed that the one-pot method achieved higher drug loading efficiency and longer release time compared to the post-loading method. In vitro experiments demonstrated that DMOG@ZIF-8 enhanced the migration, tube formation, and protein secretion related to angiogenesis in human umbilical vein endothelial cells, as well as the mineralization, enzyme activity, and protein secretion related to osteogenesis in bone marrow mesenchymal stem cells. In vivo experiments using a cranial critical defect model confirmed that the addition of OD@ZIF-8 significantly promoted vascularized bone formation. Overall, loading ZIF-8 with DMOG shows promise for reconstructing critical-sized bone defects, with the one-pot method being preferred for preparing the drug-loading system.

A Mechanically Reinforced Super Bone Glue Makes a Leap in Hard Tissue Strong Adhesion and Augmented Bone Regeneration

Shanshan Hu et al.

Summary: The innovative bone glue developed in this study meets the criteria for current bone tissue engineering strategies, providing minimized surgical trauma, stabilization of the injured area, and establishment of a dynamic osteogenic microenvironment. The glue, inspired by mussel adhesive properties, combines poly(vinyl alcohol), L-dopa amino acid, and zeolitic imidazolate framework-8 for strong adhesion to hard tissues with a maximum strength of 10 MPa. It also exhibits excellent biocompatibility, stable degradability, and in vitro osteogenic ability. Furthermore, when used in combination with Bio-Oss, it promotes sustained osteogenesis, resulting in satisfactory healing in calvarial bone defect models in rabbits.

Drug-Delivery Nanoplatform with Synergistic Regulation of Angiogenesis-Osteogenesis Coupling for Promoting Vascularized Bone Regeneration

Yahong Li et al.

Summary: A nano DFO-loaded ZIF-8 drug delivery system was successfully synthesized and demonstrated to promote angiogenesis and osteogenesis in vitro, leading to bone regeneration. RNA sequencing analysis revealed upregulation of the PI3K-AKT-MMP-2/9 and HIF-1 alpha pathways in HUVECs, resulting in the formation of new blood vessels. The mechanism by which DFO@ZIF-8 NPs promoted bone regeneration potentially involved the synergistic effect of angiogenesis-osteogenesis coupling and Zn2+-mediation of the MAPK pathway.

An efficient metal-organic framework-based drug delivery platform for synergistic antibacterial activity and osteogenesis

Yunhui Si et al.

Summary: In this study, a metal-organic framework (MOF) based drug delivery platform was used to modify titanium implants, providing them with antibacterial activity, biocompatibility, and osteogenesis-promoting properties. The modified implants released Zn2+ and MV, which caused oxidative damage to E. coli and S. aureus, inhibiting bacterial proliferation. Furthermore, the MV@ZIF-8 coating promoted osteogenic differentiation of human bone mesenchymal stem cells by activating the canonical Wnt/b-catenin signaling pathway. This research demonstrates the potential application of MOF-based drug delivery platforms in bone tissue engineering.
