The flip-flop configuration of the PABP-dimer leads to switching of the translation function

Sohyun Gu et al.

Summary: PABP is a translation initiation factor that interacts with the poly(A) tail of mRNAs, stimulating translation by interacting with eIF4G. It is present in higher quantities than mRNA and eIF4G in cells. Research showed that RNA-free PABPs dimerize in a head-to-head configuration, interfering with PABP-eIF4G interaction, and Poly(A) RNA can convert the PABP-PABP complex into a Poly(A)-PABP complex, facilitating PABP-eIF4G interaction.

PHLDB2 Mediates Cetuximab Resistance via Interacting With EGFR in Latent Metastasis of Colorectal Cancer

Maochao Luo et al.

Summary: This study identifies PHLDB2 as a key player in latent liver metastasis of colorectal cancer (CRC). Chemotherapeutic-induced oxidative stress promotes N6-methyladenosine modification of PHLDB2 messenger RNA, leading to increased protein expression of PHLDB2. Upregulated PHLDB2 stabilizes EGFR and promotes its nuclear translocation, resulting in activation of EGFR signaling and resistance to cetuximab. The study proposes PHLDB2 as a potential target for CRC treatment.

DAP5 enables main ORF translation on mRNAs with structured and uORF-containing 5' leaders

Ramona Weber et al.

Summary: This study reveals the important role of the non-canonical initiation factor DAP5 in selective translation of transcripts, particularly those with long 5' leader sequences and persistent uORF translation. These mRNAs preferentially encode signaling factors, and DAP5 is also involved in translation re-initiation.

Transcriptomic Analysis of Human Fragile X Syndrome Neurons Reveals Neurite Outgrowth Modulation by the TGFβ/BMP Pathway

Liron Kuznitsov-Yanovsky et al.

Summary: In this study, using a neural differentiation model of FX human embryonic stem cells, it was found that the absence of FMRP protein leads to delayed development and maturation of FX neurons, as well as dysregulation of genes related to the extracellular matrix and the TGF beta/BMP signaling pathway. Inhibition of the TGF beta/BMP signaling pathway can rescue the decreased neurite outgrowth in FX neurons.

The molecular biology of FMRP: new insights into fragile X syndrome

Joel D. Richter et al.

Summary: Inactivation of the gene encoding fragile X mental retardation protein (FMRP) drives the impairments in brain development and function that underlie fragile X syndrome. Richter and Zhao illustrate how innovative genetic and molecular biology tools have enhanced our understanding of both FMRP's function and the causes of fragile X syndrome pathophysiology.

Translation of ABCE1 Is Tightly Regulated by Upstream Open Reading Frames in Human Colorectal Cells

Joana Silva et al.

Summary: ABCE1 is tightly regulated by uORFs during translation, with the main coding sequence being repressed; this repression can be slightly overcome under conditions of endoplasmic reticulum stress, indicating that ABCE1 expression does not have oncogenic functions.
