LHRH conjugated gold nanoparticles assisted efficient ovarian cancer targeting evaluated via spectral photon-counting CT imaging: a proof-of-concept research

Dhiraj Kumar et al.

Summary: Emerging multifunctional nanoparticulate formulations, with nano-meter scale size and surface chemistry, can act as therapeutic delivery agents and diagnostic tools for non-invasive real-time monitoring using imaging technologies. This study evaluated the selective uptake of 18 nm and 80 nm sized gold nanoparticles (AuNPs) by ovarian cancer (OC) cells in vitro, and found that the 18 nm AuNPs were taken up 4 times more by SKOV3 cells compared to OVCAR5 cells. Luteinizing hormone-releasing hormone (LHRH) was conjugated to the surface of the AuNPs, making them potential contrast agents for novel spectral photon-counting CT imaging technology and a promising approach for targeted therapy of ovarian cancer.

MicroRNA-144-3p protects against chemotherapy-induced apoptosis of ovarian granulosa cells and activation of primordial follicles by targeting MAP3K9

Meng Liu et al.

Summary: In this study, enhanced apoptosis of granulosa cells and aberrant activation of primordial follicles were observed in a POF mouse model induced by cisplatin. MiR-144-3p was downregulated and MAP3K9 was upregulated in primary ovarian granulosa cells from POF mice. MiR-144-3p attenuated cisplatin-induced apoptosis of granulosa cells by silencing MAP3K9 expression and prevented premature primordial follicle depletion in cisplatin-induced POF mice through targeting Map3k9.

Extracellular vesicles from iPSC-MSCs alleviate chemotherapy-induced mouse ovarian damage via the ILK-PI3K/AKT pathway

Rui-Can Cao et al.

Summary: Chemotherapy can cause damage to ovarian tissues, leading to reproductive dysfunction and ovarian insufficiency. However, extracellular vesicles from induced pluripotent stem cell-derived mesenchymal stem cells have been found to restore ovarian follicle numbers and promote cell proliferation while inhibiting apoptosis in both cultured and in vivo ovaries. This therapeutic effect is achieved through the up-regulation of the ILK-PI3K/AKT pathway, possibly through the transfer of regulatory microRNAs targeting ILK pathway genes.

The use of viral vectors in vaccine development

Tatianna Travieso et al.

Summary: Vaccines are the most cost-efficient and equitable way to combat and eradicate infectious diseases. Nucleic acid vaccines, including DNA plasmid vaccines, mRNA vaccines, and recombinant viral vectors, have advantages such as inducing durable immune responses, high vaccine stability, and ease of large-scale manufacturing. In this review, we provide an overview of pre-clinical and clinical data on recombinant viral vector vaccines and discuss the advantages and limitations of different viral vector platforms.

Study protocol of RESCUE-ALS: A Phase 2, randomised, double-blind, placebo-controlled study in early symptomatic amyotrophic lateral sclerosis patients to assess bioenergetic catalysis with CNM-Au8 as a mechanism to slow disease progression

Steve Vucic et al.

Summary: Amyotrophic lateral sclerosis (ALS) is a progressive and fatal neurodegenerative disorder with limited treatment options. The RESCUE-ALS study investigates the potential of the novel nanocatalytic drug, CNM-Au8, to enhance metabolic and energetic capacity of motor neurones. This multicenter, randomized, double-blind study aims to evaluate the efficacy and safety of CNM-Au8 in ALS patients over a 36-week treatment period.

Summary: Gold nanoparticles have great potential in biomedicine but may also have toxicity, primarily affecting the liver, spleen, and kidneys. Studies show controversy, but toxicity is mainly determined by the properties and preparations of NPs, with functionalizing NP surfaces potentially regulating and detoxifying their uptake.

miRNA Targets: From Prediction Tools to Experimental Validation

Giulia Riolo et al.

Summary: MicroRNAs are post-transcriptional regulators of gene expression in animals and plants, playing gene-regulatory roles by pairing to microRNA responsive elements on target mRNAs. The identification of miRNA-mRNA target interactions is fundamental for discovering the regulatory network governed by miRNAs.
