Systematic engineering of Escherichia coli for efficient production of nicotinamide riboside from nicotinamide and 3-cyanopyridine

Summary: In this study, a whole-cell catalysis method for efficient synthesis of NR was established using E. coli. The researchers confirmed the high catalytic activity of UshA in synthesizing NR and identified the genes involved in NR degradation. They also demonstrated the important role of UshA's signal peptide in NR transport. By introducing nitrile hydratase, they achieved the conversion of 3-cyanopyridine to NR. The efficient biosynthesis of NR in E. coli was achieved with high titers using nicotinamide and 3-cyanopyridine as substrates in a 5-L bioreactor.

Solvent tolerant enzymes in extremophiles: Adaptations and applications

Summary: Nonaqueous enzymology has unique possibilities in bio-catalysis, but the presence of solvents often hinders enzyme catalysis. Information about solvent-stable enzymes is scarce, yet these enzymes have great value in biotechnology. Extremophiles, particularly those with inherent structural attributes, can be a valuable source for studying solvent-stable enzymes. Protein engineering approaches are used to enhance catalytic flexibility and stability under non-aqueous conditions. This review provides a comprehensive overview of solvent-stable enzymes from extremophilic microorganisms and discusses strategies for optimal immobilization and catalysis.

Summary: In this study, we cloned and expressed a promising enzyme Ct-PtxD for NAD(P)H regeneration from a marine cyanobacterium. Ct-PtxD showed increased resistance to salt ions and organic solvents compared to previously reported PtxD enzymes. It also demonstrated higher efficiency in the coupled chiral conversion of trimethylpyruvic acid, making it a potential candidate for industrial applications.

Structure-conditioned amino-acid couplings: How contact geometry affects pairwise sequence preferences

Summary: This study explores the relationship between a protein's sequence and its conformation and introduces pairwise potentials conditioned on the fully defined geometry of interacting backbone fragments. The results show that the structure-conditioned coupling energies accurately reflect pair preferences in different structural contexts and encode native sequence information more reliably. By clustering interaction motifs based on structure or energy, we establish a tangible link between modular sequence and structure elements. Additionally, scoring CASP models with structure-conditioned energies significantly improves correlation with structural quality compared to using a contact potential.

Substrate access tunnel engineering for improving the catalytic activity of a thermophilic nitrile hydratase toward pyridine and pyrazine nitriles

Summary: This study identified a potential gating residue, beta Leu48, in a thermophilic NHase from an extremophile, which can enhance the enzymatic activity towards various pyridine and pyrazine nitriles. Structural analysis revealed that the mutant residue could form a hydrogen bond that expands the substrate access tunnel entrance and accelerates substrate migration.
