Catalytic Asymmetric Synthesis of Axially Chiral Diaryl Ethers through Enantioselective Desymmetrization

Summary: In this study, a highly efficient catalytic asymmetric synthesis of diaryl ether atropisomers was reported via an organocatalyzed enantioselective desymmetrization protocol. The chiral phosphoric acid-catalyzed asymmetric electrophilic aromatic aminations of symmetrical substrates provided a series of diaryl ether atropisomers with excellent yields and enantioselectivities. The utilization of the 1,2-benzenediamine moiety in the products allowed for the facile construction of heterocycles, leading to a wide range of structurally diverse and novel azaarene-containing diaryl ether atropisomers.

Identification and structure-activity relationships for a series of N, N-disubstituted 2-aminobenzothiazoles as potent inhibitors of S. aureus

Summary: A collection of small molecule compounds was screened against drug-resistant bacterial pathogens, and compound 1 was found to be a potent inhibitor of Staphylococcus aureus and methicillin-resistant S. aureus. N-Heterocyclic Carbene-Catalyzed Atroposelective Synthesis of N-N Axially Chiral 3-Amino Quinazolinones

Summary: While the selective synthesis of biaryls with axially chiral C-C bonds is well-known, the synthesis of compounds containing axially chiral C-N bonds is less explored, and the construction of axially chiral N-N bonds has received little attention. This study demonstrates the selective amidation reaction catalyzed by N-heterocyclic carbenes (NHCs), leading to the atroposelective synthesis of N-N axially chiral 3-amino quinazolinones. The NHC-catalyzed reaction of quinazolinones with a free N-H group and α,β-unsaturated aldehydes under oxidative conditions provides the atropisomeric quinazolinone derivatives under mild conditions and with a broad scope. Preliminary studies on experimental and density functional theory-based determination of N-N rotational barrier are also presented.

A Dynamic Kinetic Resolution Approach to Axially Chiral Diaryl Ethers by Catalytic Atroposelective Transfer Hydrogenation

Summary: This paper presents a dynamic kinetic resolution method for the synthesis of axially chiral diaryl ethers. By using a Bronsted acid catalyzed atroposelective transfer hydrogenation (ATH) reaction, the desired diaryl ethers could be obtained in moderate to good chemical yields (up to 79%) and high enantioselectivities (up to 95% ee) under standard reaction conditions. These structural motifs are interesting precursors for further transformations and may have potential applications in the synthesis of chiral ligands or catalysts.
