Role of endoplasmic reticulum stress and autophagy in the transition from acute kidney injury to chronic kidney disease

Tahib Habshi et al.

Summary: Acute kidney injury (AKI) and chronic kidney disease (CKD) are global health concerns, and the transition from AKI to CKD requires attention from healthcare providers. This transition is a major risk factor for the development of cardiovascular disease and end-stage kidney disease. The mechanisms involving endoplasmic reticulum (ER) stress and autophagy in this AKI-to-CKD transition are still not fully understood.

Tubular Epithelial Cell HMGB1 Promotes AKI-CKD Transition by Sensitizing Cycling Tubular Cells to Oxidative Stress: A Rationale for Targeting HMGB1 during AKI Recovery

Zhi Bo Zhao et al.

Summary: Late diagnosis is a challenge for AKI treatment, but targeting AKI-CKD transition may improve outcomes. HMGB1, a nuclear regulator of transcription and a driver of necroinflammation, modulates AKI-CKD transition. Preemptive HMGB1 antagonism can attenuate AKI-CKD transition in a model of persistent kidney hypoxia.

Modulation of gentamicin-induced acute kidney injury by myo-inositol oxygenase via the ROS/ALOX-12/12-HETE/GPR31 signaling pathway

Isha Sharma et al.

Summary: In this study, a potentially novel signaling pathway in gentamicin-induced acute kidney injury worsened by overexpression of proximal tubular enzyme, MIOX, was elucidated. The results showed that MIOX-transgenic mice had severe tubular lesions associated with increased serum creatinine and proteinuria, while MIOX-KO mice had mild lesions. Furthermore, the study identified the ROS/ALOX-12/12HETE/GPR31 signaling axis as a key mechanism in this process, regulating the inflammatory response, kinase signaling cascade, and albuminuria.

Adverse Maternal and Fetal Outcomes in a Novel Experimental Model of Pregnancy after Recovery from Renal Ischemia-Reperfusion Injury

Summary: Recent clinical studies show that women with a history of acute kidney injury may experience poor maternal and fetal outcomes during pregnancy, despite fully recovering renal function prior to conception. The mechanisms behind these adverse outcomes in pregnancy following AKI remain unclear.

Regulation of Tissue Inflammation by 12-Lipoxygenases

Abhishek Kulkarni et al.

Summary: 12-LOX is a key lipid metabolizing enzyme that plays an important role in inflammatory diseases. Increased activity of 12-LOX can lead to tissue damage and enhanced migration of immune cells, while inhibition of this enzyme can protect against disease development and progression in animal models.
