Summary: This study confirmed the covalent binding of 4-hydroxybenzoic acid (4HBA) to i3-casein after ultra high temperature-like processing (UHT) in model aqueous systems. Molecular dynamics simulations showed that the covalent addition of 4HBA affected the protein density and solvent accessibility, increasing the number of hydrogen bonds but reducing their persistence. This led to decreased stability and increased flexibility of the protein.

Characterization of the improved functionality in soybean protein-proanthocyanidins conjugates prepared by the alkali treatment

Summary: This study investigated the changes in functionality, structure, and protein composition in soybean protein-proanthocyanidins (SPI-PC) conjugates prepared by the alkali method with dose-ranging PC molecular grafting. The results showed that higher PC levels led to the formation of larger polymers, altered protein structure, and changes in protein and peptide composition. The antioxidant activity and emulsifying properties of SPI-PC conjugates increased with higher PC levels, while IgE binding capacity decreased.

Examination of β-lactoglobulin-ferulic acid complexation at elevated temperature using biochemical spectroscopy, proteomics and molecular dynamics

Summary: This study investigated the molecular interactions between ferulic acid and beta-lactoglobulin under high temperature conditions. The results showed that ferulic acid altered the secondary structure of beta-lactoglobulin and formed a covalent interaction between the two components. Molecular docking studies predicted the covalent interaction between lysine and ferulic acid, forming a new adduct. Protein hydrolysis experiments confirmed the chemical nature of the interaction.

Biotinylated caffeic acid covalent binding with myofibrillar proteins in alkaline conditions: Identification of protein-phenol adducts and alterations in protein properties

Summary: The study investigated the effects of covalent interactions between myofibrillar proteins (MP) and caffeic acid (CA). Protein-phenol adducts were identified using biotinylated caffeic acid (BioC) as a substitute for CA. The total sulfhydryls and free amines content decreased, while the alpha-helix structure of MP increased. MP gel properties slightly enhanced at low CA dosages, but significantly impaired at high CA dosages. Myosin heavy chain (MHC)-BioC and Actin-BioC adducts were identified and their concentrations showed a negative correlation with gel properties.

Sea bass protein-polyphenol complex stabilized high internal phase of algal oil Pickering emulsions to stabilize astaxanthin for 3D food printing

Summary: This study demonstrated the protective effect of sea bass protein (SBP)-(-)-epigallocatechin-3-gallate (EGCG) covalent complex-stabilized high internal phase (algal oil) Pickering emulsions (HIPPEs) on astaxanthin and algal oils. The formation of the SBP-EGCG complex improved the wettability and antioxidant activity, leading to the stabilization of HIPPEs. The incorporation of SBP-EGCG complex resulted in dense particle shells surrounding the oil droplets and a network structure, providing HIPPEs with high viscoelasticity and thermal stability for 3D printing applications.

Covalent chemical modification of myofibrillar proteins to improve their gelation properties: A systematic review

Summary: Covalent modifications play a dose-dependent and dual-role in the gelation properties of myofibrillar proteins. Mild oxidation, enzyme catalysis, and isoelectric solubilization/precipitation treatments are beneficial in forming more aligned and cross-linked three-dimensional networks for myofibrillar protein gels due to moderate protein aggregation. However, excessive aggregation hinders the gelation behavior, leading to reduced gelation quality. Glycation effectively increases hydrophilicity of myofibrillar proteins, while phenolic conjugation provides MPs with novel bioactivity.
