Managing sample introduction problems in hydrophilic interaction liquid chromatography

Mark R. Taylor et al.

Summary: Sample injection in HILIC can cause issues when the injection solvent has higher elution strength than the mobile phase, resulting in asymmetric peak shapes and poor efficiency. Strategies to overcome this problem include pre-column dilution with a weak solvent and using alternative organic solvents in the injection solvent. Controlling the volume of injection solvents and using small sample volumes is recommended to reduce mismatch effects.

Strategies to circumvent the solvent strength mismatch problem in online comprehensive two-dimensional liquid chromatography

Soraya Chapel et al.

Summary: Online comprehensive two-dimensional liquid chromatography is a powerful technique for separating highly complex samples, achieving impressive peak capacities by adding a second dimension of separation. However, the online transfer of solvents can lead to severe injection effects, reducing peak capacity and intensity. Various methods have been developed to overcome these effects in recent years, with advantages and drawbacks discussed in a critical manner.

Comparison of existing strategies for keeping symmetrical peaks in on-line Hydrophilic Interaction Liquid Chromatography x Reversed-Phase Liquid Chromatography despite solvent strength mismatch

Soraya Chapel et al.

Summary: This study compared four strategies for method development in on-line HILIC x RPLC, including flow splitting, on-line dilution, ASM dilution, and Total Breakthrough Strategy. The Total Breakthrough Strategy showed the highest peak capacity while on-line dilution using ASM showed the lowest. When separating a tryptic digest in sub-hour separations, peak intensities were generally higher with on-line dilution approaches, but Total Breakthrough Strategy provided approximately 7-fold higher peak intensities compared to flow splitting.

A comprehensive study on the phenomenon of total breakthrough in liquid chromatography

Soraya Chapel et al.

Summary: The differences in elution strength between the sample solvent and the mobile phase can lead to breakthrough and total breakthrough phenomena in chromatographic separations. The presence of positive charges on the molecule is essential for observing the total breakthrough phenomenon, while factors such as injection conditions and analyte retention impact the onset of both phenomena.
