Comparison between 5 extractions methods in either plasma or serum to determine the optimal extraction and matrix combination for human metabolomics

Maryne Lepoittevin et al.

Summary: This study compared the application of five solvent-based and solid-phase extraction methods in plasma and serum, and analyzed the extracts using four LC-MS protocols. The results showed that methanol and methanol/acetonitrile precipitation had broad specificity and outstanding accuracy. There was also high orthogonality between methanol-based methods and solid-phase extraction, which could increase the metabolome coverage. However, the potential benefits need to be considered against time constraints, sample consumption, and the risk of low reproducibility of the solid-phase extraction method.

Using matrix-induced ion suppression combined with LC-MS/MS for quantification of trimethylamine-N-oxide, choline, carnitine and acetylcarnitine in dried blood spot samples

Wan-Hui Lu et al.

Summary: A method combining MIIS with LC-MS/MS was developed to estimate blood volume and quantify TMAO and its related substances in DBSs, showing high accuracy in volume estimation and quantification. The validated method demonstrated excellent linearity, precision, and accuracy, with stable analytes in DBSs for at least 30 days. The successful application of the method in real-world DBS samples shows its potential to facilitate understanding of the gut microbiota in human health.

Comparison of dried blood spot and plasma sampling for untargeted metabolomics

Nicole H. Tobin et al.

Summary: Plasma and dried blood spot (DBS) samples yield distinct metabolite profiles highly predictive of individual subjects. Following standardization, DBS and plasma metabolite profiles show strong concordance. Random forests classification models for cases versus controls exhibit similar performance in plasma and DBS.
