Aryl hydrocarbon receptor signals in epithelial cells govern the recruitment and location of Helios+ Tregs in the gut

Summary: CD4(+) Foxp3(+) regulatory T cells (Tregs) play a crucial role in the homeostasis of the colon, but the mechanism controlling their localization is still unclear. This study demonstrates that indigo naturalis (IN), an aryl hydrocarbon receptor (AhR) agonist, increases the numbers of Helios+ Tregs and MHC class II+ epithelial cells (ECs) in the colon. Additionally, IN treatment leads to an increase in CD25(+) T cells near the surface of ECs in ulcerative colitis (UC) patients compared to other therapies.

Genetic tracing reveals transcription factor Foxp3-dependent and Foxp3-independent functionality of peripherally induced Treg cells

Summary: The role of Foxp3 in peripheral pTreg cells and the mechanisms supporting their differentiation remain poorly understood. This study used genetic tracing to identify microbiota-induced pTreg cells and found that many of their distinguishing features were Foxp3 independent. While Foxp3 was critical for the suppression of certain diseases, pTreg cells could suppress colonic effector T cell expansion in a Foxp3-independent manner. Thus, Foxp3 and the tolerogenic signals preceding and promoting its expression confer distinct facets of pTreg functionality.

Microbiota imbalance induced by dietary sugar disrupts immune-mediated protection from metabolic syndrome

Summary: Intestinal microbes protect against obesity and metabolic syndrome by inducing commensal-specific Th17 cells. High-fat, high-sugar diet depletes Th17-inducing microbes and promotes metabolic disease, while recovery of commensal Th17 cells restores protection. Th17 cells regulate lipid absorption across the intestinal epithelium in an IL-17-dependent manner. Dietary sugar and the presence of Faecalibaculum rodentium displace Th17-inducing microbiota and contribute to the immuno-pathogenicity of sugar.
