Bioactive peptides for boosting stem cell culture platform: Methods and applications

Ahmed Abdal Dayem et al.

Summary: Peptides, which are short protein fragments, have the ability to mimic full-length proteins and have become a promising alternative to recombinant proteins due to their specificity, stability, low cost, and customization capabilities. They can enhance stem cell adhesion, proliferation, and differentiation and can be immobilized on culture plates or integrated into scaffolds. This review evaluates the applications of various peptides in stem cell culture and discusses the challenges and future directions for advancing peptide applications in tissue regeneration and clinical settings.

Ductal delivery of extracellular vesicles promote the recovery from salivary gland inflammation

Donghyun Kim et al.

Summary: Salivary gland dysfunction worsens the quality of life, but the treatment options are limited. This study investigates the therapeutic potential of extracellular vesicles (EVs) derived from salivary gland mesenchymal stem cells (sgMSCs). The findings demonstrate that sgMSC-EVs are more effective in promoting recovery from salivary gland inflammation and injury, suggesting the use of EVs as therapeutic tools for salivary gland dysfunction.

Stem cell-derived exosomes: emerging therapeutic opportunities for wound healing

Chuchao Zhou et al.

Summary: Wound healing is a complex process involving hemostasis, inflammation, proliferation, and tissue remodeling. Mesenchymal stem cell-derived exosomes (MSC-exos) have lower immunogenicity and high biological activity, and play a role in regulating the behavior of various skin cells. This study focuses on exploring the specific roles and mechanisms of different MSC-exos in wound healing, as well as their limitations and potential applications. Understanding the biological properties of MSC-exos is crucial for developing cell-free therapies for wound healing and cutaneous regeneration.

Thermostable Human Basic Fibroblast Growth Factor (TS-bFGF) Engineered with a Disulfide Bond Demonstrates Superior Culture Outcomes in Human Pluripotent Stem Cell

Sejong Kim et al.

Summary: Human pluripotent stem cells are a valuable resource for cell therapy products due to their ability to differentiate into various tissues. However, culturing these stem cells requires the presence of basic fibroblast growth factor (bFGF) to maintain their stemness. Unfortunately, bFGF has a short half-life and needs continuous supply, posing challenges for high-quality stem cell culture. To address this issue, a thermostable bFGF was developed, which showed better proliferation, stemness, morphology, and differentiation of human embryonic stem cells. This thermostable bFGF can play a critical role in securing high-quality stem cells during cultivation.

Summary: This article summarizes the physiologic functions, mechanistic role, and current commercial landscape of exosomes in regenerative aesthetics. Exosomes primarily exert their effects in a paracrine function and modulate the interactions between keratinocytes and other cells of the skin. They have shown therapeutic capabilities in wound healing, scar modulation, alopecia, and skin rejuvenation. However, their safety, efficacy, potency, and dosages remain to be determined through robust human clinical trials.

3D Culture and Interferon-gamma Priming Modulates Characteristics of Mesenchymal Stromal/Stem Cells by Modifying the Expression of Both Intracellular and Exosomal microRNAs

Matteo Bulati et al.

Summary: This study investigated the effects of priming on the miRNA expression of MSCs and their exosomes. The results showed that different priming conditions led to distinct miRNA expression patterns in both MSCs and exosomes, but there were common deregulated miRNAs related to tissue repair/regeneration. The findings suggest that both 3D culture and IFN-gamma treatment can enhance the therapeutic potential of MSCs by modulating miRNA expression. Overall, this study provides valuable insights into the molecular mechanisms underlying the miRNA-mediated beneficial effects of MSCs.

Mesenchymal stromal/stem cells spheroid culture effect on the therapeutic efficacy of these cells and their exosomes: A new strategy to overcome cell therapy limitations

Ali Hazrati et al.

Summary: Cell therapy, specifically mesenchymal stem/stromal cell (MSCs) transplantation, has emerged as a new treatment method with therapeutic efficacy proven in clinical applications. MSCs exert their therapeutic functions through various mechanisms such as differentiation potential, immunomodulatory properties, production of therapeutic exosomes, growth factors and cytokines, and anti-apoptotic effects. However, limitations in exosome production and therapeutic ability have prompted researchers to explore methods that enhance the therapeutic potential of exosomes, such as MSCs cultured in spheroids. This review discusses the enhanced properties of MSCs and their exosomes in spheroid culture.

Kwonwoo Song et al.

Summary: Interstitial cystitis/bladder pain syndrome (IC/BPS) is a chronic disease characterized by severe pelvic pain. Research has found that mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) have enhanced wound healing and anti-inflammatory capabilities, and show promise in treating IC/BPS. The increased level of TGF-p1 in these EVs may contribute to their anti-inflammatory activity.
