Comparative responses of flocculating and nonflocculating yeasts to cell density and chemical stress in lactic acid fermentation

Engineering Flocculation for Improved Tolerance and Production of d-Lactic Acid in Pichia pastoris

Summary: In this study, the tolerance to d-lactic acid and production were improved in Pichia pastoris by introducing a flocculation gene from Saccharomyces cerevisiae (ScFLO1) and a d-lactate dehydrogenase gene from Leuconostoc pseudomesenteroides (LpDLDH). The engineered strain KM71-ScFlo1-LpDLDH achieved a d-lactic acid titer of 5.12 +/- 0.35 g/L in 48 h, which was 2.6-fold higher than the control strain. Transcriptomics analysis revealed the upregulations of genes involved in lactate transport and iron metabolism as the mechanism of increased tolerance to d-lactic acid.

Enhanced surface display efficiency of β-glucosidase in Saccharomyces cerevisiae by disruption of cell wall protein-encoding genes YGP1 and CWP2

Summary: This study explored the effects of two cell wall protein genes, CWP2 and YGP1, on beta-glucosidase activity on the yeast cell surface. The disruption of YGP1 and CWP2 increased beta-glucosidase activity, and further transcriptional analysis revealed changes in the expression of genes related to cell wall structure, biogenesis, and integrity.

The flocculant Saccharomyces cerevisiae strain gains robustness via alteration of the cell wall hydrophobicity

Summary: This study explores the resistance of Saccharomyces yeast strains to inhibitory chemical compounds (ICCs) released from lignocellulosic biomass. The F118 strain of S. cerevisiae shows robustness and unique flocculation trait under ICC stress. Gene expression analysis reveals that the Mot3p gene plays a key role in the response to ICC concentration. Swapping the Mot3p gene enhances cell wall hydrophobicity and response to ICC stress, resulting in faster fermentation and higher ethanol production.
