More than a Virulence Factor: Patulin Is a Non-Host-Specific Toxin that Inhibits Postharvest Phytopathogens and Requires Efflux for Penicillium Tolerance

Holly P. Bartholomew et al.

Summary: Mycotoxin contamination is a significant factor contributing to food spoilage and waste worldwide. The mycotoxin patulin, produced by Penicillium spp. during postharvest fruit decay, has harmful effects on human health and is resistant to pasteurization and fermentation. There is limited knowledge about the impact of patulin on postharvest fruit-pathogen interactions. This study found that patulin application on apple cultivars reproduced blue mold symptoms that were independent of cultivar type and dose-dependent. Similar symptoms were also observed in pear and mandarin orange. Exposure of Penicillium isolates to exogenous patulin resulted in delayed germination but viable colonies were still produced. However, four common postharvest phytopathogenic fungi were completely inhibited by patulin during conidial germination, suggesting the toxin's importance in Penicillium dominating the postharvest environment. The use of a broad-spectrum efflux pump inhibitor demonstrated the role of efflux in Penicillium's resistance to patulin during conidial germination. This study provides new insights into patulin auto-resistance, its mode of action, and its inhibitory role in fungal-fungal interactions, which can contribute to the development of toxin and decay mitigation strategies.

Study on the biocontrol effect and physiological mechanism of Hannaella sinensis on the blue mold decay of apples

Rouling Lin et al.

Summary: In this study, a yeast strain, Hannaella sinensis, was selected for its good control effect on blue mold decay in apples. The research aimed to investigate the biocontrol effect of H. sinensis on blue mold decay in apples and the underlying mechanisms. The results demonstrated that H. sinensis effectively controlled blue mold decay in apples and inhibited the germination of spores and growth of germ tubes of P. expansum. Furthermore, H. sinensis colonized apple wounds and surfaces, showing its environmental adaptability and ability to compete for resources with other microbiota. Additionally, H. sinensis induced the activities of resistance-related enzymes and the expression of corresponding genes in apples, leading to enhanced disease resistance. Overall, H. sinensis showed great potential as an antagonistic yeast for the prevention and treatment of postharvest blue mold decay in apples.

Comparative Penicillium spp. Transcriptomics: Conserved Pathways and Processes Revealed in Ungerminated Conidia and during Postharvest Apple Fruit Decay

Holly P. Bartholomew et al.

Summary: This study investigated the gene expression during apple fruit decay caused by Penicillium spp. using transcriptomics. It identified differentially expressed genes between two Penicillium species with different levels of aggressiveness and revealed unique genes expressed during apple decay. Gene ontology analysis showed differential regulation in fungal transport and metabolism genes, suggesting a flux in nutrient acquisition and detoxification strategies. Additionally, the study discovered pre-loaded transcripts in conidia from blue mold fungi and revealed the expression dynamics of known fungal virulence factors during apple decay.

The Good, the Bad, and the Ugly: Mycotoxin Production During Postharvest Decay and Their Influence on Tritrophic Host-Pathogen-Microbe Interactions

Holly P. Bartholomew et al.

Summary: Understanding the impact of various toxins on the beneficial and harmful microorganisms within the host microbiome is essential and can lead to the identification of safe analogs for antimicrobial activity to reduce fruit decay. Exploring the composition of the microbial carposphere of host plants may help in developing a microbial consortium to maintain quality during storage and reduce mycotoxin contamination.

Genomic Analyses of Penicillium Species Have Revealed Patulin and Citrinin Gene Clusters and Novel Loci Involved in Oxylipin Production

Guohua Yin et al.

Summary: Blue mold of apple is caused by various Penicillium species, with P. expansum being the most aggressive while P. solitum is weak in infecting apple fruit. Genomic analyses of different Penicillium species showed differences in mycotoxin production and toxin emitting capabilities. The study provides insights into the genetic bases of mycotoxins and toxic volatile organic compounds in Penicillium spp.
