Two Birds One Stone Strategy for the Site-Specific Analysis of Core Fucosylation and O-GlcNAcylation

Summary: Here, a two birds one stone strategy has been described for the site-specific analysis of core fucosylation and O-GlcNAcylation using mutant endoglycosidases and a glycan probe. A temperature-sensitive polymer was introduced for the enrichment of labeled glycopeptides, which can be enzymatically released for mass spectrometry analysis. This strategy allows simultaneous profiling of core-fucosylated and O-GlcNAcylated glycoproteomes from complex samples.

Chemoproteomic mapping of the glycolytic targetome in cancer cells

Summary: Hyperactivated glycolysis is a metabolic characteristic of cancer cells, and glycolytic metabolites have nonmetabolic functions as signaling molecules. However, how these metabolites interact with their targets remains unclear. This study introduces a new approach called TRAP to identify target candidates and interactions of glycolytic metabolites in cancer cells, revealing diverse regulatory modalities and potential for cancer therapy.

An electroaffinity labelling platform for chemoproteomic-based target identification

Quantitative Chemoproteomic Profiling with Data-Independent Acquisition-Based Mass Spectrometry

Summary: Activity-based protein profiling (ABPP) is a powerful tool for annotating protein functions and discovering targets of bioactive ligands. In this study, we combined data-independent acquisition (DIA) MS with ABPP to develop a label-free quantitative chemical proteomic method called DIA-ABPP. With good reproducibility and high accuracy, DIA-ABPP allows for high-throughput quantification, enabling comprehensive profiling of functional proteomes and interactions with bioactive small molecules.

Liquidambaric acid inhibits Wnt/β-catenin signaling and colon cancer via targeting TNF receptor-associated factor 2

Summary: This study reveals that liquidambaric acid (LDA) can inhibit oncogenic Wnt/beta-catenin signaling by targeting tumor necrosis factor receptor-associated factor 2 (TRAF2). TRAF2 plays a crucial role in the formation of TRAF2/beta-cateniniTCF4/TNIK complex in colon cancer cells, and this interaction is disrupted in the presence of LDA. These findings highlight the significance of TRAF2 in Wnt/beta-catenin signaling and colon cancer, and suggest that TRAF2 could be a potential target for cancer therapy.

Summary: Bile acids are endogenous metabolites with important functions. Through chemoproteomic methods, we identified the histidine kinase EnvZ as a novel sensor for bile acids in bacteria. Activation of the downstream signaling pathway mediated by EnvZ helps bacteria tolerate bile acids. This discovery opens new opportunities to combat bile acid-tolerating pathogens.

CETSA interaction proteomics define specific RNA-modification pathways as key components of fluorouracil-based cancer drug cytotoxicity

Summary: The lack of detailed understanding of the mechanism of action of many cancer drugs is hindering their optimal use. This study used a high-resolution implementation of the proteome-wide cellular thermal shift assay to investigate the protein interaction changes induced by the antimetabolite 5-fluorouracil (5-FU) and related nucleosides. The findings revealed the expected effects on the known main target and enzymes in specific pathways, but also identified novel protein interactions and pathways associated with 5-FU, including RNA-modification and-processing pathways. The study further highlighted key components of the 5-FU mechanism of action, indicating that the inhibition of a specific target is not necessary for cell proliferation.

Thermal proteome profiling identifies the membrane- bound purinergic receptor P2X4 as a target of the autophagy inhibitor indophagolin
