Gut virome-colonising Orthohepadnavirus genus is associated with ulcerative colitis pathogenesis and induces intestinal inflammation in vivo

Luca Massimino et al.

Summary: This study found that the virome-associated protein encoded by the hepatitis B virus (HBx) is detected in about 45% of patients with ulcerative colitis (UC) and is associated with UC progression. Experimental findings showed that HBx induces colonic inflammation and its silencing reverses the colitis phenotype. HBx acts as a transcriptional regulator in epithelial cells, leading to barrier leakage and altering mucosal immunity. This study provides a new perspective on targeted treatments for the virome composition of the gut microbiota.

The GSTP1/MAPKs/BIM/SMAC modulatory actions of nitazoxanide: Bioinformatics and experimental evidence in subcutaneous solid Ehrlich carcinoma-inoculated mice

Samar Imbaby et al.

Summary: This study aimed to explore novel chemotherapeutic targets and evaluated the therapeutic effects of Nitazoxanide (NTZ) in Ehrlich ascites carcinoma and subcutaneous inoculated solid tumour form (SEC). The results showed that NTZ inhibited tumor growth and metastasis and induced apoptosis in tumor cells by modulating the MAPKs, Glutathione-S-transferase P1 (GSTP1), and SMAC signaling pathways. Therefore, NTZ may be a potential therapeutic approach for solid tumors.

The immunomodulatory effects of probiotics and azithromycin in dextran sodium sulfate-induced ulcerative colitis in rats via TLR4-NF-xB and p38-MAPK pathway

Shereen E. Elkholy et al.

Summary: Ulcerative colitis (UC) is a chronic autoimmune disease characterized by its relapsing and remitting nature, and it is considered a major healthcare problem. This study evaluated the therapeutic effects of oral probiotics and azithromycin regimens in a rat model of UC, and found that the combination therapy showed the most synergistic beneficial effects in improving histological architecture and reducing inflammation markers in UC tissues. These findings highlight the potential of combining probiotics and azithromycin as a therapeutic approach to enhance the quality of life in UC patients.

Palmitoyltransferase ZDHHC3 Aggravates Nonalcoholic Steatohepatitis by Targeting S-Palmitoylated IRHOM2

Minxuan Xu et al.

Summary: Underestimation of the complexity of NASH pathogenesis hinders the development of new drugs and targeted therapies. This study revealed that IRHOM2 is post-translationally S-palmitoylated at C476, which facilitates its translocation and stabilization. ZDHHC3 was identified as the key acetyltransferase required for IRHOM2 palmitoylation, and its increased association with IRHOM2 promotes palmitoylation and prevents ubiquitination-mediated degradation.

Colorectal Cancer in Inflammatory Bowel Disease: Mechanisms and Management

Shailja C. Shah et al.

Summary: Patients with inflammatory bowel disease (IBD) have an increased risk of developing colorectal cancer (CRC). Chronic inflammation plays a key role in the development of neoplastic progression, resulting in dysplastic precursor lesions. Colitis-associated CRC shares molecular similarities with sporadic CRC and the microbiome and host immune system may also play a role. However, further research is needed to understand the specific molecular differences and their role in inflammation-associated cancers.

Old and New Players of Inflammation and Their Relationship With Cancer Development

Rodolfo Chavez-Dominguez et al.

Summary: This text discusses how pathogens or genotoxic agents continuously affect the human body, triggering acute inflammatory reactions for efficient elimination of harmful substances. Chronic inflammation, if sustained, can lead to cancer development by increasing genomic instability and creating a tumor-promoting environment. The immune system plays a crucial role in eliminating transformed cells, but excessive inflammation may amplify local damage and hinder immune response.
