Designing Tailored Thiosemicarbazones with Bespoke Properties: The Styrene Moiety Imparts Potent Activity, Inhibits Heme Center Oxidation, and Results in a Novel Stealth Zinc(II) Complex?

Mahendiran Dharmasivam et al.

Summary: A novel and selective antitumor agent, 3-thiosemicarbazone (PPP44mT), and its analogues with distinctive properties were synthesized and characterized. The inclusion of a styrene moiety prevented the oxidation of oxy-myoglobin or oxy-hemoglobin, which is different from other potent thiosemicarbazones such as DpC or Dp44mT. PPP44mT and its Cu(II) complexes exhibited immediate cytotoxicity against SK-N-MC tumor cells, while [Zn(PPP44mT)2] showed delayed antiproliferative efficacy and superior inhibition of cyclin D1 expression compared to DpC or Dp44mT.

Innovative thiosemicarbazones that induce multi-modal mechanisms to down-regulate estrogen-, progesterone-, androgen- and prolactin-receptors in breast cancer

Faten Shehadeh-Tout et al.

Summary: This study investigates a new class of anti-cancer agents for the treatment of ER-positive breast cancer. These agents inhibit multiple growth factor receptors and down-stream signaling, leading to decreased expression of hormone receptors and key resistance pathways. In vivo experiments demonstrate that these agents effectively inhibit ER-positive breast cancer growth. This represents an innovative non-hormonal, multi-modal therapeutic approach.

Human serum albumin as a copper source for anticancer thiosemicarbazones

Martin Schaier et al.

Summary: Thiosemicarbazones (TSCs) are biologically active compounds that have potential anticancer activity by chelating iron and targeting ribonucleotide reductase. Recent findings suggest that the stability and reduction properties of copper complexes are essential for the action of specific TSC derivatives. This study investigated the interaction between TSCs and human serum albumin (HSA) as a potential source of copper, and found that TSCs specifically chelate copper from HSA and form ternary HSA conjugates.

Copper-Catalyzed Glutathione Oxidation is Accelerated by the Anticancer Thiosemicarbazone Dp44mT and Further Boosted at Lower pH

Enrico Falcone et al.

Summary: The study found that Cu-II catalyzes the oxidation of glutathione (GSH) faster in the presence of the anticancer thiosemicarbazone Dp44mT, especially at acidic pH. Density functional theory (DFT) calculations, as well as electrochemical and spectroscopic studies, revealed the mechanism behind this acceleration.

Competition between reactive adsorption desulfurization and olefin hydrogenation over the NiO/ZnO-Al2O3-SiO2 adsorbent

Feng Ju et al.

Summary: Reactive adsorption desulfurization (RADS) is effective for upgrading fluidized catalytic cracking of gasoline by achieving high desulfurization activity and low olefin saturation. The temperature has a significant impact on the desulfurization and olefin saturation reactions in RADS. Competitive adsorption between sulfides and olefins occurs at the active sites, but the amount of sulfides adsorbed is higher than that of olefins, indicating that olefin hydrogenation has limited influence on the desulfurization reaction. Increasing the Ni loading can improve desulfurization performance but also intensify olefin hydrogenation.

Breaking the cycle: Targeting of NDRG1 to inhibit bi-directional oncogenic cross-talk between pancreatic cancer and stroma

Bekesho Geleta et al.

Summary: The study reveals that NDRG1 plays an inhibitory role in the interaction between PaCa and PSCs, reducing signaling pathways like HGF and IGF-1 and inhibiting metastasis and cell migration in pancreatic cancer. DpC, a novel anticancer agent, upregulates NDRG1 expression to decrease PaCa cells' sensitivity to ligands secreted by PSCs.

Innovative therapies for neuroblastoma: The surprisingly potent role of iron chelation in up-regulating metastasis and tumor suppressors and down-regulating the key oncogene, N-myc

Tharushi P. Wijesinghe et al.

Summary: Iron is essential for biological processes in cancer cells, with their higher demand making them susceptible to iron depletion. Iron chelation therapy, particularly using Desferrioxamine (DFO), has shown effective anti-proliferative activity against various cancer types, including neuroblastoma, by down-regulating the oncogene N-myc. DFO has limitations such as poor membrane permeability and short half-life, leading to the development of metal-binding ligands like di-2-pyridylketone thiosemicarbazone (DpT) series, with compounds like DpC showing better anti-proliferative activity against neuroblastoma tumors in vivo without cardiotoxicity.
