Summary: Stimulus-responsive nanomaterials have shown promise in controllable drug delivery systems for cancer therapy. In this study, iron (III) carboxylate metal-organic framework nanoparticles coated with glycyr- rhetinic acid-chitosan conjugate were successfully fabricated as a pH-responsive and target-selective system for delivering doxorubicin to hepatocellular carcinoma cells. The nanocarriers exhibited uniform size, high drug loading efficiency, and pH-dependent controlled drug release. In vitro experiments demonstrated their excellent inhibitory effects on cancer cells and targeted delivery ability. The results suggest that these nanoparticles have potential applications in cancer therapy as a pH-responsive controlled drug delivery system.

Summary: A magnetic liposome-based dual-targeting delivery system was designed for the treatment of idiopathic pulmonary fibrosis (IPF), effectively delivering dexamethasone (Dex) into M2 macrophages in the lung to inhibit the progression of IPF. The mannose-modified magnetic nanoparticles were loaded on the surface of liposomes, allowing recognition and internalization by M2 macrophages in situ under the influence of a static magnetic field (SMF). The reduction of M2 macrophages and inhibition of their polarization led to a decrease in α-SMA expression and collagen deposition, demonstrating the effectiveness of this novel therapeutic system for IPF treatment.

Summary: In this study, a strategy to block the redox systems and enhance ferroptotic cancer cell death was reported using a layered double hydroxide (LDH) nanoplatform. The nanoplatform, siR/IONs@LDH, can efficiently release iron oxide nanoparticles (IONs) and DHODH inhibitor (siR) in a pH-responsive manner, generate toxic reactive oxygen species (ROS) via a Fenton reaction, and synergistically induce cancer cell death by promoting lipid peroxidation accumulation. In vivo therapeutic evaluations demonstrated excellent tumor growth inhibition by the nanomedicine without detectable side effects. This study provides new insights into nanomaterial-mediated tumor ferroptosis therapy.

Summary: This research proposes the one-pot preparation of polydopamine (PDA) decorated mesoporous silica nanoparticle (PMSN) for the thermal and tumor micro-environment (TME) responsive colorectal tumor therapy. The PMSN pores were utilized for loading Fe3+. Lauric acid (LA) was selected as a phase-change ligand to coat the surface of Fe3+-loaded PMSN and prevent the leakage of Fe3+. Bovine serum albumin (BSA) was chosen as a stabilizer to provide colloidal stability for the PMSN-Fe-LA-BSA nanopartilces (PMFLB). Under near infrared laser, the light-sensitive PDA generated heat to kill colorectal cancer cells via hyperthermia. The heat induced the phase-change of LA and triggered the release of Fe3+, which reacted with endogenous H2S in the colorectal TME. The transformed Fe3+ into Fe2+ then initiated the Fenton reaction with H2O2, generating hydroxyl radical (•OH). The Fe2+ was converted back to Fe3+, which further reacted with H2S to enhance the chemodynamic therapy of colorectal tumor. The thermosensitive PMFLB that operates in synergy with the colorectal TME offers a new approach for the rational design of multifunctional nano-therapeutic agents.

In this study, ultra-small-sized gemcitabine-based nanoparticles were combined with folic acid conjugation for enhanced dual targeted delivery to tumor cells and tumor-associated macrophages. The co-delivery system showed small particle size and achieved significant reduction of tumor growth in a breast cancer model.
