Summary: This study retrospectively analyzed the association between tumor mutational burden (TMB) and/or the density of stromal CD8-positive tumor-infiltrating lymphocytes (CD8(+)TILs) and clinical factors in patients with squamous cell carcinoma of the uterine cervix treated with radiotherapy. The results showed no association between TMB and CD8(+)TIL density, but an involvement of ARID1A mutations in antitumor immune responses.

Summary: This study investigated the density of tumor-infiltrating lymphocytes (TILs) and tertiary lymphoid structures (TLS) in squamous cell laryngeal carcinomas (LSCC) and their correlation with tumor pathology. The results showed a significant decrease in TIL and TLS densities in the tumor areas. Additionally, low densities of TILs and TLS were associated with aggressive behavior.

Prognostic and Predictive Relevance of Tumor-Infiltrating Lymphocytes in Squamous Cell Head-Neck Cancer Patients Treated with Radical Radiotherapy/Chemo-Radiotherapy

Summary: Microenvironmental conditions play a crucial role in controlling the entrance and thriving of cytotoxic lymphocytes in tumors. In this study, the density of tumor-infiltrating lymphocytes (TILs) in squamous cell head-neck tumors (HNSCC) was investigated, and the link between markers of hypoxia and TIL density was assessed. The results showed that high TIL density was associated with better survival outcomes, and overexpression of HIF1 alpha, HIF2 alpha, and CA9 was significantly linked with poor infiltration by TILs. These findings suggest that targeting HIF and CA9-related pathways in immunologically cold tumors may be a potential therapeutic intervention.

Simultaneous integrated boost concepts in definitive radiation therapy for esophageal cancer: outcomes and toxicity

Summary: This retrospective analysis demonstrates that utilizing a simultaneous integrated boost (SIB) concept in definitive (chemo)radiation therapy is safe and feasible, with acceptable outcomes in this patient cohort. The SIB radiation therapy may serve as potential partial compensation for omitted simultaneous chemotherapy, particularly in elderly patients not eligible for chemotherapy. Prospective studies are needed for validation of these findings.

Summary: This study investigated the curative effect and prognostic factors of concurrent chemoradiotherapy for esophageal cancer using SIB-IMRT. The results indicated that T-stage, N-stage, clinical stage, short-term efficacy, and concurrent chemotherapy were important prognostic factors for survival. Single chemotherapy concurrent chemoradiotherapy should be considered in the management of esophageal cancer due to its lower toxicity and comparable effectiveness to double chemotherapy.
