On the Physical Stability of Leucine-Containing Spray-Dried Powders for Respiratory Drug Delivery

However, the presence of leucine fibers during stability has been observed. In this study, the origin of these fibers was determined to be the presence of amorphous or less stable crystalline polymorphs of leucine, but they did not affect the stability or aerosol performance of the powders.

Effect of Amphotericin B on the Thermodynamic Properties and Surface Morphology of the Pulmonary Surfactant Model Monolayer during Respiration

Summary: During the COVID-19 pandemic, the treatment of pulmonary fungal infection faced challenges. Amphotericin B has potential as an inhalation treatment for pulmonary fungal infections, but its clinical use is limited due to renal toxicity. In this study, the interaction between amphotericin B and the pulmonary surfactant monolayer was investigated. Results showed that at lower molar ratios of AmB to lipids, attractive intermolecular forces were observed, while at higher molar ratios, repulsive forces were predominant. These findings provide insights into the interaction between the pulmonary surfactant and different doses of amphotericin B during respiration.

Identifying genetic variants associated with amphotericin B (AMB) resistance in Aspergillus fumigatus via k-mer-based GWAS

Summary: In this study, a k-mer-based genome-wide association study (GWAS) was performed to analyze the relationship between genetic variants and amphotericin B resistance in A. fumigatus. The study found that indel variants are more strongly associated with amphotericin B resistance compared to SNP sites. Enrichment analysis suggested that sphingolipid synthesis and transmembrane transport may play a role in the resistance of A. fumigatus to amphotericin B. These findings contribute to the identification of candidate genetic markers for amphotericin B resistance screening in A. fumigatus.

Summary: We tested the efficacy of different antifungal drugs against invasive moulds collected worldwide from 2018 to 2021. Most Aspergillus isolates remained susceptible to the tested drugs, but resistance to azoles was increasing in both North America and Europe, especially in A. fumigatus. Amphotericin B and caspofungin showed potential activity against azole-resistant A. fumigatus and posaconazole and amphotericin B were most effective against Mucorales.

Tuning the aggregation state of amphotericin B formulations is a promising strategy to achieve a more selective response against fungal cells and minimize toxicity in mammalian cells.

Summary: This study aimed to address the limited therapeutic effect of poly-L-lysine (PLL) on pulmonary infection due to its hygroscopicity by co-spray-drying PLL with L-leucine (LL) to achieve moisture-resistance and desirable aerosolization performance. Optimal PLL:LL ratio was determined to obtain particles with different characteristics. The developed PLL dry powder inhalations (DPIs) exhibited superior anti-hygroscopicity, aerosolization performance, and rheological properties. In addition, the DPIs demonstrated stability under high humidity conditions and showed promising results in treating pulmonary infections, as confirmed by in vitro and in vivo experiments.
