Self-Healing Injectable Hydrogels for Tissue Regeneration

Pascal Bertsch et al.

Summary: Biomaterials that can self-heal and maintain their structural integrity have significant advantages in biomedicine. Self-healing injectable hydrogels, a new type of biomaterial, have emerged in the past decade. These hydrogels can temporarily fluidize and then recover their mechanical properties under shear stress. They can be administered locally and minimally invasively, and have moldable properties for personalized medicine. Additionally, they facilitate tissue regeneration through their viscoelastic and diffusive nature. However, challenges remain in balancing self-healing and injectability with physical stability, establishing consensus on rheological characterization, and translating them into therapeutically effective formulations for specific tissues.

Bioactive composite Janus nanofibrous membranes loading Ciprofloxacin and Astaxanthin for enhanced healing of full-thickness skin defect wounds

Wei Zhang et al.

Summary: In this study, skin-inspired multilayer Janus composite nanofibrous membranes were designed and prepared as wound dressings with superhydrophilicity and superhydrophobicity. The membranes were investigated for their wettability, morphology, in-vitro release, cytotoxicity, antimicrobial activity, and healing effect. Results from animal experiments and histological analysis demonstrated that the composite membranes were more effective in improving the healing of full-thickness defect wounds and promoting the growth of collagen fibers and collagen deposition. The composite membranes consisted of a drug-loading layer with a superhydrophilic surface for self-pumping and drugs backflow, a hydrophobic polyvinylidene fluoride layer for delaying exudates loss, and a superhydrophobic coating with self-cleaning properties for avoiding the adhesion of foreign microorganisms.

A biodegradable pH and glutathione dual-triggered drug delivery system based on mesoporous silica, carboxymethyl chitosan and oxidized pullulan

Qian Rui et al.

Summary: A simple and smart drug controlled delivery system was developed by using biodegradable mesoporous silica nanoparticles (BMSN) for the loading of an anti-cancer drug methotrexate (MTX). The MTX loaded BMSN (BMSN-MTX) was encapsulated in the hydrogels of carboxymethyl chitosan (CMCS)/oxidized pullulan (OPL) generated through Schiff base reaction. The release of MTX from the BMSN-MTX-gel can be triggered by both pH and glutathione (GSH), making it an efficient drug delivery system.
