Domain motions, dimerization, and membrane interactions of the murine guanylate binding protein 2

Summary: This study focuses on the role of mGBP2 in controlling Toxoplasma gondii replication by recruiting to and destroying the parasitophorous vacuole membrane. Lipid-binding assays confirm the specific affinity of mGBP2 for cardiolipin, and fluorescence microscopy reveals its interaction with giant unilamellar vesicles. Molecular dynamics simulations show that mGBP2 exhibits a large-scale hinge motion when bound to a cardiolipin-containing membrane, potentially leading to membrane destruction by forming supramolecular complexes.
