Tuning ultrasmall theranostic nanoparticles for MRI contrast and radiation dose amplification

Needa Brown et al.

Summary: In this work, second-generation AGuIX (R) nanoparticles (AGuIX-Bi) were synthesized and validated, showing enhanced radiation dose amplification by replacing some Gd3+ cations with Bi3+ in the nanoparticles. The safety and efficacy of AGuIX-Bi were confirmed in vitro and in vivo, with improved DNA damage and a significant delay in tumor growth.

Use of magnetic resonance image-guided radiotherapy for breast cancer: a scoping review

Alexandra Berlangieri et al.

Summary: This scoping review examined the clinical implementation of MRI-guided radiotherapy (MRIgRT) in breast cancer patients, finding that accelerated partial breast irradiation (APBI) is the most common treatment form. It also highlighted the importance of using bolus in specific areas to protect skin and prevent unwanted doses. Overall, APBI treatment on MR-linac is deemed feasible.

AGuIX nanoparticles enhance ionizing radiation-induced ferroptosis on tumor cells by targeting the NRF2-GPX4 signaling pathway

Hao Sun et al.

Summary: This study demonstrated the effectiveness of AGuIX nanoparticles in radio-sensitization of triple-negative breast cancer. It increased DNA damage level by compromising the homologous recombination repair pathway and may regulate the anti-ferroptosis system by inhibiting the NRF2-GSH-GPX4 signaling pathway.

Gadolinium-based ultra-small nanoparticles augment radiotherapy-induced T-cell response to synergize with checkpoint blockade immunotherapy

Huijuan Song et al.

Summary: This study developed a synergistic strategy based on AGuIX-mediated radiotherapy and immune checkpoint blockade to improve B16 tumor therapy. AGuIX nanoparticles exacerbated radiation-induced DNA damage and induced immunogenic cell death of B16 tumor cells, activating immune responses. Combining AGuIX-mediated radiotherapy with programmed cell death protein 1 blockade showed excellent therapeutic effects.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Genistein inhibits radiation-induced invasion and migration of glioblastoma cells by blocking the DNA-PKcs/Akt2/Rac1 signaling pathway

Xiongxiong Liu et al.

Summary: This study demonstrated that genistein can effectively inhibit radiation-induced invasion and migration of GBM cells by specifically binding to DNA-PKcs and blocking its pathway. DNA-PKcs is not only a potential target for cancer therapy, but also a reliable biomarker for predicting radiation-induced invasion and migration of GBM cells.

Therapeutic Efficacy of Carbon Ion Irradiation Enhanced by 11-MUA-Capped Gold Nanoparticles: An in vitro and in vivo Study

Pengcheng Zhang et al.

Summary: The study demonstrated that 11-mercaptoundecanoic acid-coated gold nanoparticles (mAuNPs) showed radiosensitivity to carbon ion irradiation in in vivo and in vitro experiments. The radiosensitization effect on mice tumor could be achieved by activating the mitochondrial apoptosis pathway and increasing tumor tissue apoptosis. This is the first in vivo evidence for radiosensitization of mAuNPs in tumor-bearing mice exposed to carbon ion irradiation.
