Effect of Early vs Standard Approach to Tracheostomy on Functional Outcome at 6 Months Among Patients With Severe Stroke Receiving Mechanical Ventilation The SETPOINT2 Randomized Clinical Trial

Julian Bosel et al.

Summary: This study aimed to test whether early tracheostomy improved functional outcome among patients with stroke receiving mechanical ventilation. The results showed that there was no significant difference in the rate of survival without severe disability at 6 months between the early tracheostomy group and the standard tracheostomy group.

Quantification of hematoma and perihematomal edema volumes in intracerebral hemorrhage study: Design considerations in an artificial intelligence validation (QUANTUM) study

Natasha Ironside et al.

Summary: This article describes a study that aims to apply a fully automated algorithm to quantify hematoma and perihematomal edema volumes in intracerebral hemorrhage. The study utilizes an equivalence hypothesis testing paradigm to directly assess the equivalence of the automated and semi-automated quantification methods with the manual quantification method. The challenges encountered in the design of this study may have implications for translating artificial intelligence medical technology into clinical practice.

Aprepitant attenuates NLRC4-dependent neuronal pyroptosis via NK1R/PKCδ pathway in a mouse model of intracerebral hemorrhage

Peng Jin et al.

Summary: This study found that inhibition of NK1R receptor using Aprepitant can attenuate neuronal pyroptosis and improve neurological function after intracerebral hemorrhage (ICH). The neuroprotective effects of Aprepitant may be mediated by inhibition of NK1R/PKC delta signaling pathways.

Neutrophil-to-lymphocyte ratio predicts in-hospital mortality in intracerebral hemorrhage

Li Li et al.

Summary: This study aimed to investigate the potential of neutrophil-to-lymphocyte ratio as a predictor of in-hospital mortality in patients with severe intracerebral hemorrhage. The results showed that a higher neutrophil-to-lymphocyte ratio was associated with an increased risk of in-hospital death, indicating its potential as a biomarker for outcome prediction.

Deep learning shows good reliability for automatic segmentation and volume measurement of brain hemorrhage, intraventricular extension, and peripheral edema

Xianjing Zhao et al.

Summary: The deep learning model based on no-new-Net showed good reliability in automated segmentation and volumetric measurements of intracerebral hemorrhage (ICH), intraventricular extension of intracerebral hemorrhage (IVH), and perihematomal edema (PHE) in primary ICH. The model achieved high Dice scores, sensitivities, and positive predictive values for lesion segmentation, and demonstrated excellent concordance for ICH and IVH, and good concordance for PHE between manual and automated volume measurements. The model was able to provide automatic segmentation and volume analysis for each patient in approximately 15 seconds.

Mesenchymal Stem Cells Transplantation in Intracerebral Hemorrhage: Application and Challenges

Yu-hua Gong et al.

Summary: Intracerebral hemorrhage (ICH) is a leading cause of death and long-term disability worldwide. Research has shown that mesenchymal stem cell (MSC) therapy can improve neuronal defects caused by ICH, but key issues regarding the survival, differentiation, and safety of grafted MSCs still need to be resolved for clinical translation.

NLRP3 Inflammasome Assembly in Neutrophils Is Supported by PAD4 and Promotes NETosis Under Sterile Conditions

Patrick Muenzer et al.

Summary: The canonical NLRP3 inflammasome participates in NETosis under sterile conditions by regulating NLRP3 and ASC protein levels, while genetic ablation of NLRP3 signaling impairs NET formation. Inhibition of NLRP3 decreases NETosis, and NLRP3 deficiency results in reduced NET density in thrombi in a mouse model of deep vein thrombosis.
