Synthesis and Evaluation of Europium Complexes that Switch on Luminescence in Lysosomes of Living Cells

Summary: A set of four luminescent Eu-III complexes with an extended aryl-alkynylpyridine chromophore have been studied, showing significant differences in pH-dependent behavior in their absorption and emission spectral response. The best-behaved probe exhibited no significant emission dependence on the concentration of endogenous cations, reductants, and serum albumin, and was able to monitor time-dependent lysosomal acidification effectively. Through optimization of the 'switch-on' intensity ratio, enhancement factors between 250 to 1330 were measured between pH 8 and 4.
