Modular Engineering of DNAzyme-Based Sensors for Spatioselective Imaging of Metal Ions in Mitochondria

Summary: DNAzyme-based biosensor technology is developed for spatiotemporally controlled imaging of metal ions in mitochondria, overcoming the limitation of lack of subcellular precision. This technology allows for mitochondria-specific imaging of Zn2+ in living cells using optically activatable DNAzyme sensor and upconversion nanoparticles. The system is also applied for monitoring mitochondrial Zn2+ during drug treatment and assessing dynamic Zn2+ change and pharmacological interventions in cellular models. This method paves the way for investigating the pathophysiological roles of metal ions at the subcellular level using engineered DNAzyme sensors.

A Fluid Multivalent Magnetic Interface for High-Performance Isolation and Proteomic Profiling of Tumor-Derived Extracellular Vesicles

Summary: The study develops a microfluidic chip named FluidmagFace for efficient isolation, release, and protein profiling of tumor-derived extracellular vesicles (T-EVs). The FluidmagFace increases affinity by 105-fold and improves isolation efficiency by 13.9% compared to a non-fluid interface. It minimizes contamination and increases sensitivity by two orders of magnitude. With the chip, T-EVs were detected in all tested cancer samples and differentially expressed proteins were identified compared to healthy controls. The FluidmagFace opens a new avenue for cancer diagnosis and biomarker discovery.

Advanced Pt hollow nanospheres/rubrene nanoleaves coupled with M-shaped DNA walker for ultrasensitive electrochemiluminescence bioassay

Summary: In this study, an intense electrochemiluminescence (ECL) system was developed using Pt hollow nanospheres/rubrene nanoleaves (Pt HNSs/Rub NLs) for ultrasensitive detection of carcinoembryonic antigen (CEA). The M-shaped DNA walker (M-DNA walker) was employed as a signal switch. The Pt HNSs exhibited superior catalytic performance and pore confinement-enhanced ECL, amplifying the ECL intensity of Rub NLs/dissolved O2 (DO) binary system. The M-DNA walker and tracks were confined on the Pt HNSs, promoting reaction efficiency and enhancing specificity for CEA recognition.

Upconversion Luminescence-Initiated and GSH-Responsive Self-Driven DNA Motor for Automatic Operation in Living Cells and In Vivo
