Glucocorticoid-driven mitochondrial damage stimulates Tau pathology

Fang Du et al.

Summary: Du et al. identify a mechanism by which glucocorticoid stress hormones induce mitochondrial damage and tau pathology in mice, and show that therapeutic targeting of mitochondria may be a promising strategy for reducing stress-related neurotoxicity and cognitive impairment.

Mitochondrial dysfunction and oxidative stress in Alzheimer's disease, and Parkinson's disease, Huntington's disease and Amyotrophic Lateral Sclerosis -An updated review

Taha Alqahtani et al.

Summary: Misfolded proteins in the central nervous system can induce oxidative damage, impacting mitochondrial function and leading to neurodegenerative diseases such as Alzheimer's, Parkinson's, Huntington's and Amyotrophic Lateral Sclerosis. Mitochondrial dynamics play a crucial role in cellular apoptosis and are affected by these diseases. The dysfunction of mitochondria contributes to the selective neurodegeneration seen in these disorders. Novel techniques are being explored to treat mitochondrial malfunction and oxidative stress in these neurodegenerative diseases.

Understanding How Physical Exercise Improves Alzheimer's Disease: Cholinergic and Monoaminergic Systems

Boyi Zong et al.

Summary: Alzheimer's disease is an age-related neurodegenerative disorder characterized by protein accumulation and neuronal damage. Dysregulation of cholinergic and monoaminergic systems is involved in the pathogenesis. Exercise therapy has the potential to prevent pathological features and improve cognitive function by modulating neurotransmitter activity.

Peroxisomal Proliferator-Activated Receptor β/δ Deficiency Induces Cognitive Alterations

Triana Espinosa-Jimenez et al.

Summary: Peroxisome proliferator-activated receptor beta/delta (PPAR beta/delta) plays a key role in microglial homeostasis and metabolism in the central nervous system. However, its contribution to neuronal and memory processes is still not well-understood. This study aimed to evaluate the role of PPAR beta/delta in neuropathological pathways associated with memory impairment and the modulation of these pathways by obesity. The results showed that PPAR beta/delta deficiency affects neuronal and synaptic structure, leading to memory dysfunction.

Transcription Factor TWIST1 Integrates Dendritic Remodeling and Chronic Stress to Promote Depressive-like Behaviors

Jin-Gang He et al.

Summary: The study found that the expression of TWIST1 in the brain is positively correlated with depressive behaviors, chronic stress can elevate TWIST1 expression in the medial prefrontal cortex of mice which can be reversed by fluoxetine treatment. Overexpression of TWIST1 increases susceptibility to stress, while knockdown of TWIST1 prevents defective dendrite morphogenesis in mice and alleviates depressive-like behaviors. The prodepressant property of TWIST1 is mediated by the repression of miR-214-PPAR-delta signaling and mitochondrial function, which can be mimicked by genetic and pharmacological inhibition of PPAR-delta.

The role of peroxisome proliferator-activated receptors (PPAR) in immune responses

Anthos Christofides et al.

Summary: PPARs are fatty acid-activated transcription factors that regulate energy metabolism, with three subtypes identified: PPAR alpha, PPAR gamma, and PPAR beta/delta. They play distinct roles in oxidative and adipose tissues, as well as in immune cells, contributing to immune cell differentiation and inflammation.

Excess tau PET ligand retention in elderly patients with major depressive disorder

Summary: The study found that tau protein accumulations in the brains of MDD patients were significantly higher than healthy controls, especially in patients with psychotic symptoms. These findings suggest that tau depositions may underlie MDD, particularly in those with psychotic symptoms.

Melatonin ameliorates cognitive deficits through improving mitophagy in a mouse model of Alzheimer's disease

Chongyang Chen et al.

Summary: The study shows that melatonin treatment can improve mitophagy in Alzheimer's disease, restoring mitochondrial function, reducing Aβ pathology, and improving cognition. Concurrent treatment with chloroquine and melatonin inhibits the positive effects of melatonin alone.

Depression in Alzheimer's Disease: A Delphi Consensus on Etiology, Risk Factors, and Clinical Management

Luis Aguera-Ortiz et al.

Summary: This study gathered expert opinions on the management of dementia and depression in elderly patients, highlighting the importance of routine cognitive check-ups in depressed individuals, careful interpretation of neuropsychological assessments, selection of appropriate antidepressants, and consideration of exercise and psychological interventions alongside pharmacological treatment. The study also emphasized the need for further research and specific recommendations for this comorbid patient population.

A High Fat/Cholesterol Diet Recapitulates Some Alzheimer's Disease-Like Features in Mice: Focus on Hippocampal Mitochondrial Dysfunction

Gianni Mancini et al.

Summary: The study found that a high fat/cholesterol diet may lead to mitochondrial dysfunction and significantly decreased nitric oxide production in a mouse model of Alzheimer's disease, exacerbating brain tissue damage. In normal mice, the diet only caused moderate hypercholesterolemia but resulted in significant decline in spatial memory.
