However, the key determinants and analytical methods for monitoring and predicting protein instability at these interfaces remain difficult to identify. This review discusses the mechanisms, potential harmful materials, mitigation strategies, and current methods for studying surface-induced protein instability, aiming to aid early-stage screening and formulation development of biologics.

The contamination of pharmaceutical products with N-nitrosamines can result from multiple factors including API synthesis, contaminated solvents and water, degradation during formulation processing or storage, as well as contaminated excipients and printing inks. This review aims to comprehensively compile and analyze the reported and plausible sources of nitrosamine impurities in pharmaceutical products, providing valuable insights into the complex landscape of N-nitrosamine contamination.

A Novel Method for Monitoring N-Nitrosamines Impurities Using NH2-MIL-101(Fe) Mediated Dispersive Micro-Solid Phase Extraction Coupled with LC-MS/MS in Biopharmaceuticals

Summary: A highly efficient and convenient method using an amine-functionalized metal-organic framework as sorbent coupled with LC-MS/MS was developed for the simultaneous determination of 12 N-nitrosamines in biopharmaceuticals. The method was validated and showed good linearity, repeatability, and precision. The proposed adsorption mechanism on NH2-MIL-101(Fe) was supported by FTIR spectra. The method was successfully applied for the detection of trace nitrosamines in biopharmaceuticals.

Impact of Extractables/Leachables from Filter Materials on the Stability of Protein-Based Pharmaceutical Products

Summary: This study evaluates the extractable and leachable (E/L) substances from different polymeric filter membranes and their potential effects on protein stability. The results show that polyamide, polycarbonate, and polyethersulfone filters have higher E/L content compared to polytetrafluoroethylene and regenerative cellulose filters. The E/L substances from regenerative cellulose and polyethersulfone are surface-active and alter the particle size/structure, resulting in protein aggregation. Additionally, bisphenol A is found to be released from polycarbonate filters and could pose health risks when administered with pharmaceutical products.
