Summary: This study aimed to investigate the impact of carnosic acid (CA) on indomethacin (IND)-induced gastric ulceration. The results showed that CA pretreatment attenuated both gross morphological lesions and histopathological alterations induced by IND, and it achieved gastroprotective effects by reducing oxidative stress and suppressing inflammation.

Summary: This research evaluated the cardioprotective properties of chlorogenic acid (CGA) in doxorubicin (DOX)-induced cardiotoxicity and identified one of the possible mechanisms for its protective effects. The study found that CGA ameliorated oxidative stress and cardiomyocyte apoptosis by upregulating the Nrf2/HO-1-dependent pathway and downregulating dityrosine (DT) expression. These findings suggest that CGA may be cardioprotective, particularly in patients receiving DOX-based chemotherapy.

Summary: Osteoarthritis is a common chronic whole-joint disease, and the secretion of extracellular vesicles plays a role in its pathogenesis. Extracellular vesicles derived from mesenchymal stem cells or platelet-rich plasma have high therapeutic value for treating osteoarthritis and can be modified or incorporated into biomaterials for enhanced targeting and prolonged retention. By analyzing pathological biomarkers associated with osteoarthritis, we discuss current diagnostic and therapeutic methods and propose the potential of using extracellular vesicles for the diagnosis and treatment of osteoarthritis in the future.

Summary: Osteoarthritis is a common joint disease in the elderly and is treated through surgery, drug therapy, and exercise therapy. Exercise can alleviate the pathological changes of osteoarthritis by affecting degradation of ECM, apoptosis, inflammatory response, autophagy, and changes of ncRNA. Different types and intensities of exercise have different effects on osteoarthritis patients, and suitable exercise prescriptions are recommended.

Summary: This study proposes a novel strategy for drug delivery and treatment of osteoarthritis (OA) by using opsonized nanoparticles to attract and regulate macrophage polarization. The opsonized nanoparticles effectively promoted macrophage polarization toward the anti-inflammatory phenotype and protected nearby chondrocytes, resulting in enhanced therapeutic outcomes for OA.

Summary: This study explored the effects of natural compounds, berbamine, bergapten, and carveol, on neuroinflammatory pain associated with paclitaxel. The results showed that these compounds attenuated thermal hypersensitivity, improved motor function, normalized body weight changes, and increased the pain threshold. They also demonstrated neuroprotective effects by regulating molecular pathways.

Summary: Using single-cell RNA sequencing, this study investigated the dynamics of immune cell accumulation in the mouse knee joint after ACL rupture. Multiple immune cell types were identified, with monocytes and macrophages showing the most dramatic changes. The study also characterized different subtypes of macrophages and identified genes induced or repressed after ACL injury. These findings provide new insight into the immune microenvironment in post-traumatic osteoarthritis.

Summary: Obesity is the most important risk factor for the development and progression of osteoarthritis, and it was previously believed to be caused by excessive weight leading to joint overloading and cartilage destruction. However, recent studies have identified other factors such as adipose deposition, insulin resistance, and the improper coordination of innate and adaptive immune responses that contribute to obesity-associated osteoarthritis. Multiple inflammatory cells are recruited into the synovial joint and play an important role in pathological changes. Macrophage polarization and mediators produced by macrophages have been extensively studied and linked to the inflammatory and destructive responses in the synovium and cartilage. The role of other innate immune cells and adaptive immune cells in the pathogenesis of osteoarthritis is still under exploration.

Summary: In recent years, extracellular vesicles (EVs) secreted by mesenchymal stem cells (MSCs) have shown potential as a cell-free therapy for osteoarthritis (OA). This study investigated the effects of EVs released by cytokine-primed umbilical cord-derived MSCs (UCMSCs) on osteoarthritic chondrocyte physiology. The results showed that EVs from cytokine-primed UCMSCs could enhance chondrocyte migration and potentially induce chondrocyte regeneration. These findings suggest that cytokine priming of UCMSCs can selectively deliver therapeutic effects of EV treatment in OA and chondrocyte-related disorders.

Summary: In recent years, there have been interesting advancements in the understanding of the basis of osteoarthritis (OA). Research suggests that cartilage degeneration is preceded by subchondral bone lesions, indicating a key role of this mechanism in the pathogenesis and progression of OA. Chronic low-grade inflammation of the synovial lining, along with immunological and metabolic factors, is now considered crucial in driving inflammation and tissue destruction in OA. Additionally, the role of neuroinflammation and central sensitization mechanisms as underlying causes of chronic pain has been characterized.

Summary: Histiocytoses are rare disorders characterized by infiltration of various organs by myeloid cells, with diagnosis often delayed due to low incidence and diverse clinical manifestations. Somatic mutations in genes activating the MAP kinase pathway are commonly found in biopsy specimens, and targeted therapies with BRAF or MEK inhibitors have revolutionized treatment but challenges such as duration and prevention of neurological sequelae remain.
