The role of histone deacetylases in embryonic development

Priyanka Dsilva et al.

Summary: The basic units of chromatin are nucleosomes, consisting of DNA wrapped around histone cores. Histone lysine residue is a common site for posttranslational modifications, with acetylation being prevalent. Histone acetyltransferases (HATs/KATs) and histone deacetylases (HDACs/KDACs) regulate histone acetylation, crucial for gene expression control. Different HDAC/KDAC isoforms play significant roles in embryo development and neurogenesis, particularly in cardiac muscle differentiation. This review article summarizes various pathways and genes associated with embryonic development, the impact of HDAC/KDAC activity during stem cell differentiation and embryogenesis, and the role of different HDAC/KDAC inhibitors.

Protective Effects of Antioxidants on Cyclophosphamide-Induced Ovarian Toxicity

Ricassio S. Barberino et al.

Summary: The common limitation of anticancer chemotherapy is the damage to normal cells, and cyclophosphamide, one of the most widely used alkylating agents, can cause ovarian insufficiency and infertility. This article reviews the mechanisms of cyclophosphamide-induced ovarian damage and describes the cytoprotective compounds used to ameliorate it.

Investigation of He's Yang Chao recipe against oxidative stress-related mitophagy and pyroptosis to improve ovarian function

Chenyun Miao et al.

Summary: This study investigated the molecular mechanisms underlying the protective effects of He's Yang Chao Recipe (HSYC) on granulosa cell injury in primary ovarian insufficiency (POI). HSYC protected the ovaries from cyclophosphamide-induced damage and oxidative stress in mice with POI. The mechanism of action involved inhibiting PINK1-Parkin mitophagy and NLRP3 inflammasome activation.

Fecal transplant from myostatin deletion pigs positively impacts the gut-muscle axis

Zhao-Bo Luo et al.

Summary: The host genome and intestinal microbiota have mutual influences on each other. Deletion of the myostatin (MSTN) gene in pigs positively regulates the expression of tight junction-related genes in the intestine, leading to changes in the structure of the intestinal microbiota. Transplantation of the intestinal microbiota from MSTN-deficient pigs into mice resulted in increased muscle growth and higher levels of short-chain fatty acids.

The future of fertility preservation for women treated with chemotherapy

Lauren R. Alesi et al.

Summary: Cytotoxic chemotherapies have significant adverse effects on fertility and endocrine health, causing irreversible ovarian damage and follicle depletion. Current strategies for fertility preservation, such as oocyte and embryo freezing, do not prevent chemotherapy-induced ovarian damage effectively, nor does hormone replacement therapy fully restore endocrine function.

The Dominant Mechanism of Cyclophosphamide-Induced Damage to Ovarian Reserve: Premature Activation or Apoptosis of Primordial Follicles?

Qin Xie et al.

Summary: Cyclophosphamide (CPM) exhibits high gonadal toxicity in females and damages ovarian reserve by premature activation of primordial follicles. However, recent studies suggest that DNA double-strand breaks and apoptosis occur in primordial oocytes under CPM-induced ovarian damage. This review discusses the controversial premature activation theory and apoptosis mechanism to understand CPM's injury on ovarian reserve and aid in the development of effective ovarian protectants.

TrkB agonist antibody ameliorates fertility deficits in aged and cyclophosphamide-induced premature ovarian failure model mice

Xunsi Qin et al.

Summary: The agonistic antibody targeting the BDNF receptor TrkB can promote follicle development and oocyte maturation, reverse ovarian deficits and infertility in models of premature ovarian failure. The antibody activates TrkB signaling and restores the number and quality of oocytes and enhances fertility. This study identifies TrkB as a potential target for treating premature ovarian failure.

Epigenetic reprogramming in cloned mouse embryos following treatment with DNA methyltransferase and histone deacetylase inhibitors

Maryam Zarei et al.

Summary: This study examined the effects of the DNA methyltransferase inhibitor RG108 and the histone deacetylase inhibitor SAHA on the reprogramming parameters of cloned mouse embryos. It was found that both RG108 and SAHA separately improved the rate and quality of cloned mouse embryos, but the effects were not observed when used in combination. Furthermore, RG108 corrected the aberrant expression of key developmental genes in cloned embryos.

Coordination of zygotic genome activation entry and exit by H3K4me3 and H3K27me3 in porcine early embryos

Guowei Bu et al.

Summary: This study profiles the global H3K4me3 and H3K27me3 dynamics in porcine oocytes and embryos, revealing two previously unknown epigenetic reprogramming modes associated with zygotic genome activation (ZGA). It demonstrates the crucial role of histone modifications in regulating ZGA and suggests the potential use of porcine embryos as a model for human embryo research.

Linoleic acid reduces apoptosis via NF-kB during the in vitro development of induced parthenogenic porcine embryos

Dong-Kyung Lee et al.

Summary: Fatty acid, particularly linoleic acid, plays a crucial role in porcine preimplantation embryo development. NF-kB has been identified as an important factor in this process, mediating apoptosis pathway. The concentration of linoleic acid affects embryo development, with a positive effect at a certain concentration but developmental failure at higher concentrations. The expression level of NF-kB increases with the concentration of linoleic acid, and it is also involved in the regulation of pro-apoptotic gene expression and nuclear translocation of C-JUN.

A Tremendous Reorganization Journey for the 3D Chromatin Structure from Gametes to Embryos

Zhenping Chen et al.

Summary: The 3D chromatin structure plays a crucial role in gene expression regulation and embryonic development. Recent advancements in low-input chromatin conformation capture technologies have allowed for the study of 3D chromatin structures in gametes, zygotes, and early embryos in various species. The reorganization of 3D chromatin structure during embryogenesis shows both conserved and divergent patterns across different species, with distinct molecular mechanisms. These findings contribute to our understanding of the contribution of 3D chromatin structure reorganization to embryonic development and its role in acquiring totipotent developmental potential.

Characterizing the effects of in utero valproic acid exposure on NF-κB signaling in CD-1 mouse embryos during neural tube closure

Sidra Shafique et al.

Summary: This study demonstrated that in utero exposure to a teratogenic dose of VPA significantly downregulated mRNA levels of p65, Pim-1, and p105/p50 in CD-1 mouse embryos. Additionally, p65 protein expression was significantly upregulated in non-exencephalic embryos while p50 protein levels were significantly downregulated in both non-exencephalic and exencephalic groups at GD13. Furthermore, there were no changes in p65 and p50 protein levels in the nuclear extracts of VPA-exposed GD13 embryo heads.
