Adult stem cell niches for tissue homeostasis

Giuliana Mannino et al.

Summary: Adult stem cells are essential for tissue homeostasis, growth, and regeneration, residing in specialized environments called niches. Interactions within the niche, including intrinsic factors, supporting cells, extracellular matrix, and signaling pathways, govern cell fate and regulate proliferation and differentiation. Tissues can be classified based on their turnover rate and repair abilities, with stem cell niches playing a crucial role in tissue maintenance and response to injury or disease.

Damage-induced regeneration of the intestinal stem cell pool through enteroblast mitosis in the Drosophila midgut

Aiguo Tian et al.

Summary: In the Drosophila intestine, infection with pathogenic bacteria induces enteroblasts (ISC progenies) to enter the mitotic cycle through upregulation of the epidermal growth factor receptor (EGFR)-Ras signaling pathway. The regenerative ISCs are produced through mitosis of enteroblasts, which do not gain ISC identity before dividing.

D-Chiro-Inositol extends the lifespan of male Drosophila melanogaster better than D-Pinitol through insulin signaling and autophagy pathways

Xiaomei Du et al.

Summary: D-Chiro-Inositol (DCI) and D-Pinitol (DP) have positive effects on lifespan and antioxidant activity in Drosophila melanogaster. DCI shows slightly better effects than DP. The prolongation of lifespan by DCI and DP is achieved through regulating intestinal homeostasis and inhibiting abnormal proliferation of intestinal stem cells. Insulin signaling pathway and autophagy pathway are involved in the longevity-promoting effects of DCI and DP.

Vinculin recruitment to a-catenin halts the differentiation and maturation of enterocyte progenitors to maintain homeostasis of the Drosophila intestine

Jerome Bohere et al.

Summary: The study investigates the role of vinculin in regulating cell fate decisions in the Drosophila intestinal epithelium. It is found that vinculin prevents precocious activation and differentiation of intestinal progenitors into absorptive cells by functioning at cadherin adhesion sites. Vinculin is required at adherens junctions to maintain enteroblasts in a quiescent state and inhibit further divisions of stem cells. Manipulating cell tension demonstrates that vinculin recruitment at adherens junctions regulates enteroblast activation and cell numbers.

An amuse-bouche of stem cell regulation: Underlying principles and mechanisms from adult Drosophila intestinal stem cells

Benjamin Boumard et al.

Summary: Stem cells play essential roles in organ development and maintenance, and understanding the mechanisms by which tissue-specific stem cells renew tissues is a key goal of regenerative medicine. The Drosophila midgut provides a widely used model system for studying regulatory mechanisms of stem cell biology.

Non-apoptotic enteroblast-specific role of the initiator caspase Dronc for development and homeostasis of the Drosophila intestine

Jillian L. Lindblad et al.

Summary: The initiator caspase Dronc in Drosophila is essential for normal development of the intestine, with specific non-apoptotic functions in enteroblasts. Silencing Dronc in enteroblasts triggers hyperplasia and differentiation defects in adult intestines. These findings highlight the critical role of Dronc in controlling cell lineage and maintaining homeostasis in the adult midgut.

Neuroglian regulates Drosophila intestinal stem cell proliferation through enhanced signaling via the epidermal growth factor receptor

Martino Resnik-Docampo et al.

Summary: The SJ protein Nrg plays a crucial role in regulating ISC proliferation in the Drosophila intestine, impacting stem cell behavior and EGFR activation. This work has implications for treating intestinal disorders related to altered ISC behavior, as Nrg function is highly conserved in mammalian systems.
