Analytical isotachophoresis 1967-2022: From standard analytical technique to universal on-line concentration tool

Summary: Analytical isotachophoresis is a well-established electrophoretic separation method that allows for the analysis of mixtures and trace analysis. It has both concentrating and separating capabilities and can be used with complex-matrix samples. Recent applications have shown interest in its concentrating properties and its incorporation into multistage analysis and micropreparative procedures. This review highlights the continual development and contributions of isotachophoresis in contemporary analytical chemistry.

Preparative separation of immunoglobulins from bovine colostrum by continuous divergent-flow electrophoresis

Summary: Imaging capillary isoelectric focusing (icIEF) technology is the gold standard for monitoring monoclonal antibody (mAb) charge heterogeneity. However, traditional icIEF cannot be used in conjunction with mass spectrometry (MS) and has limitations in characterizing protein heterogeneity in depth. In this study, an integrated icIEF platform was developed that allows for rapid and reliable monitoring of protein heterogeneity, as well as MS coupling and fraction collection for further characterization.

Fractionation and online mass spectrometry based on imaged capillary isoelectric focusing (icIEF) for characterizing charge heterogeneity of therapeutic antibody

Summary: In this study, imaged capillary isoelectric focusing (icIEF) technology was used for the characterization of charge heterogeneity in a therapeutic monoclonal antibody. The icIEF-MS coupling technique was successfully applied to identify the main protein component and its four charge variants. Preparative fractionation was used to collect the major acidic and basic charge variants, which were confirmed by LC-MS at the intact protein level.

Electromembrane extraction (EME): Fundamentals and applications

Summary: Conventional methods of sample preparation have drawbacks such as high solvent and sample consumption and lengthy operation time, which are unfriendly to the environment. The miniaturization and automation of these methods, such as electromembrane extraction (EME), can reduce the use of organic solvents and provide less environmentally harmful methodologies. EME has shown great potential in the preconcentration and clean-up of complex matrices, but its performance requires a deeper understanding to improve analytical performance. This review aims to provide an overview of the fundamentals and current applications of EME extraction, which can inspire the development of new and advanced methods.

Novel chip-based isoelectric focusing device for fractionation of bacteria prior to their mass spectrometry identification

Native Separation and Metallation Analysis of SOD1 Protein from the Human Central Nervous System: a Methodological Workflow

Summary: A new methodology was developed to analyze the metal content of copper-zinc superoxide dismutase (SOD1) in tissues from the human central nervous system, demonstrating successful preservation of metal, activity, and enrichment of the protein. Results showed Cu and Zn bound to SOD1 in a ratio very close to the expected value of 1, in tissues from healthy individuals. This approach can be adapted to study other metalloproteins in human tissues and other sources.
