Self-assembled DNA nanoparticles enable cascade circuits for mRNA detection and imaging in living cells

Summary: This study develops DNA nanoparticles with GSH-responsive controllable release of hairpin reactants for the analysis and imaging of low-abundance target mRNA in living cells. The DNA nanoparticles exhibit stability, cell-specific penetration, and precise controllability. By combining multi-amplifiers and programmable DNA nanostructures, the strategy enables accurately triggered release of hairpin reactants, achieving sensitive imaging and quantitative evaluation of survivin mRNA in carcinoma cells.

MicroRNA Imaging Encounters Rolling Circle Amplification: Intracellular Na plus -Fueled Linear Programmable DNAzyme Nanostructure

Summary: A highly loaded Na+-fueled linear programmable DNAzyme nanostructure (LPDN) was proposed for sensitive detection of intracellular miRNAs. The LPDN, composed of long, single-strand DNA produced by rolling circle amplification reactions, can control the position of the DNAzyme and substrate precisely for optimal efficiency. This method enables in situ real-time imaging and evaluation of intracellular miRNA level changes, making it a promising candidate for early cancer diagnosis and imaging analysis.

Corn-Based Fluorescent Light-Up Biosensors with Improved Signal-to-Background Ratio for Label-Free Detection of Long Noncoding RNAs

Summary: In this study, a fluorescent biosensor with a low background was developed for label-free detection of long noncoding RNAs (lncRNAs). The biosensor utilized duplex-specific nuclease (DSN)-assisted target recycling amplification and transcription-driven synthesis of fluorogenic RNA aptamer-Corns. The biosensor demonstrated high sensitivity for quantifying lncRNA, capable of detecting as low as 31.98 aM. It could also accurately quantify lncRNA at the single-cell level and distinguish tumor cells from normal cells, offering potential for molecular diagnosis.

Genetically encoded RNA-based sensors with Pepper fluorogenic aptamer

DNA-Templated Bioorthogonal Reactions via Catalytic Hairpin Assembly for Precise RNA Imaging in Live Cells
