Hepatic-targeted delivery of astaxanthin for enhanced scavenging free radical scavenge and preventing mitochondrial depolarization

Summary: In this study, liver-targeted nanocarriers were designed and prepared to efficiently deliver AST by modifying it with lactobionic acid. The results showed that the lactobionic acid-modified AST nanoparticles had significant cellular uptake, and displayed excellent antioxidant properties in preventing mitochondrial depolarization. Moreover, these modified nanoparticles accumulated in the liver due to specific binding with lactobionic acid receptors, suggesting their potential use in developing functional food for liver disease nutrition intervention.

Summary: By constructing cauliflower-like carriers (CCs), the biocompatibility and stability of astaxanthin were enhanced, and the CCs exhibited targeted delivery properties. In vitro and in vivo studies demonstrated that the astaxanthin-loaded CCs improved internalization, inhibited oxidative stress-induced reactive oxygen species production and mitochondrial depolarization, relieved colitis, suppressed inflammation factors expression, and increased the abundance of beneficial bacteria in the intestine.

Hepatic macrophage targeted siRNA lipid nanoparticles treat non-alcoholic steatohepatitis

Summary: In this study, a mannose-modified HMGB1-siRNA loaded stable nucleic acid lipid particle delivery system was constructed to target liver macrophages and treat NASH. The results showed that the delivery system effectively regulated the phenotype of liver macrophages, reduced hepatic inflammation and steatosis, and restored liver function in NASH model mice. Additionally, co-administration of DHA was found to be effective in NASH treatment.
