Bumetanide Rescues Aquaporin-4 Depolarization via Suppressing p-Dys-troglycan Cleavage and Provides Neuroprotection in Rat Retinal Ischemia-Reperfusion Injury

Chunyan Chen et al.

Summary: This study investigated the effects of p-DG cleavage on AQP4 and the roles of BU in a rat retinal I/R injury model. The results showed that p-DG cleavage led to the depolarization of AQP4, while BU could recover AQP4 polarization, suppress AQP4 protein expression, attenuate retinal cytotoxic edema, and regulate the expression of p-DG and AQP4 genes. Additionally, BU also inhibited glial responses and mitochondria-mediated apoptotic protein expression, showing potential neuroprotective efficacy.

Astrocyte Endfeet in Brain Function and Pathology: Open Questions

Blanca Diaz-Castro et al.

Summary: Astrocyte endfeet surround the entire vascular tree in the central nervous system and play important roles in regulating the blood-brain barrier, cerebral blood flow, nutrient uptake, and waste clearance. They contain specialized organelles and proteins necessary for astrocyte-vascular interactions. Many neurological diseases are associated with abnormalities in astrocyte endfeet, including loss of protein polarization, vascular dysregulation, and disruption of the blood-brain barrier. This review provides an overview of astrocyte endfeet and highlights the need for future research to address gaps in our understanding.

Electroacupuncture modulates gut microbiota in mice: A potential target in postoperative cognitive dysfunction

Binbin Zhu et al.

Summary: This study investigated whether electroacupuncture (EA) can alleviate postoperative cognitive dysfunction (POCD) by modulating gut microbial dysbiosis. The results showed that EA treatment improved learning and spatial memory deficits in POCD patients and regulated the changes in intestinal flora. Additionally, EA treatment also relieved blood-brain barrier and intestinal barrier dysfunction, thus reducing hippocampal inflammation.

Reduced inhibitory and excitatory input onto parvalbumin interneurons mediated by perineuronal net might contribute to cognitive impairments in a mouse model of sepsis-associated encephalopathy

Ling Zhang et al.

Summary: This study reveals that active MMP-9 mediates PNN remodeling, leading to reduced synaptic input and abnormal gamma oscillations in hippocampal CA1, ultimately contributing to cognitive impairments in SAE induced by LPS injection. Treatment with the MMP-9 inhibitor SB-3CT can rescue most of these abnormalities.

Neuroinflammation-mediated mitochondrial dysregulation involved in postoperative cognitive dysfunction

Yan Yang et al.

Summary: Research found that activation of Drp1 and mitochondrial fragmentation increased in the hippocampus of mice after surgery and primary neurons after TNF exposure. Pretreatment with Drp1 inhibitor Mdivi-1 reduced mitochondrial fragmentation, while Drp1 knockdown blocked TNF-induced fragmentation in SH-SY5Y cells. However, Mdivi-1 negatively impacted mitochondrial function and neurite growth in primary neurons.

Summary: Our study found changes in PVS-related MRI parameters in MCI and AD, possibly due to impairment of the glymphatic system. We also revealed associations between MRI parameters and A beta deposition, neuronal change, and cognitive impairments in AD.

Limonium tetragonum Promotes Running Endurance in Mice through Mitochondrial Biogenesis and Oxidative Fiber Formation

Yong Gyun Lee et al.

Summary: The study investigated the effects of Limonium tetragonum, cultivated in a smart-farming system with LED lamps, on exercise capacity in mice. The results showed that supplementation with Limonium tetragonum significantly increased running distance and improved mitochondrial biosynthesis and the formation of oxidative slow-twitch fibers. These findings suggest a positive effect of Limonium tetragonum on enhancing endurance exercise capacity.

Disassembly and Mislocalization of AQP4 in Incipient Scar Formation after Experimental Stroke

Shervin Banitalebi et al.

Summary: This study investigates the mechanisms of scar formation in the brain and finds pronounced changes in the astrocytic protein AQP4 in the infarct border zone, which may be related to the downregulation of the AQP4 isoform AQP4ex. These findings provide new insights into the molecular basis of scar formation.

Intravenous versus Volatile Anesthetic Effects on Postoperative Cognition in Elderly Patients Undergoing Laparoscopic Abdominal Surgery A Multicenter, Randomized Trial

Yujuan Li et al.

Summary: The choice of anesthetic between propofol and sevoflurane did not show a significant difference in the incidence of delayed neurocognitive recovery 5 to 7 days after laparoscopic abdominal surgery. A high blood interleukin-6 concentration after surgical incision may be an independent risk factor for delayed neurocognitive recovery.

Notoginsenoside R1 intervenes degradation and redistribution of tight junctions to ameliorate blood-brain barrier permeability by Caveolin-1/MMP2/9 pathway after acute ischemic stroke

Bowen Liu et al.

Summary: Notoginsenoside R1 treatment significantly reduced BBB leakage and cerebral infarction volume, weakened neurological deficits in post-stroke rats. Moreover, it inhibited the activity of MMPs in infarcted cortex and striatum, down-regulated MMP2, 9 and up-regulated zo1 and claudin5 expressions in penumbra. In vitro, Notoginsenoside R1 treatment decreased OGD-induced endothelial barrier permeability, restored expressions of zo1, claudin5 on cellular membrane and cytoplasm, as well as mediated membrane redistribution of occludin and caveolin1 from actin cytoskeletal fraction.
