BPDE exposure promotes trophoblast cell pyroptosis and induces miscarriage by up-regulating lnc-HZ14/ZBP1/NLRP3 axis

Rong Wang et al.

Summary: In this study, it was found that BPDE exposure induced pyroptosis in human trophoblast cells by up-regulating the NLRP3/Caspase1/GSDMD pathway. Additionally, the high expression of lnc-HZ14 was observed in BPDE-exposed trophoblast cells and villous tissues from patients with recurrent miscarriage (RM). Lnc-HZ14 promoted trophoblast cell pyroptosis through various mechanisms including the enhancement of ZBP1/NLRP3 protein interactions.

Gestational 1-nitropyrene exposure causes anxiety-like behavior partially by altering hippocampal epigenetic reprogramming of synaptic plasticity in male adult offspring

Bo Wang et al.

Summary: The study found that gestational exposure to 1-NP caused anxiety-like behavior in male adult offspring and impaired dendritic arborization, dendritic length, and dendritic spine density in the ventral hippocampus. Gephyrin-related genes, Nrg1 and Erbb4, were also decreased in 1-NP-exposed fetuses, along with decreased TET activity and α-KG content in the fetal forebrain. Supplementation with α-KG ameliorated the effects of 1-NP, improving synaptic plasticity and anxiety-like behavior in male adult offspring.

Suppression of FPR2 expression inhibits inflammation in preeclampsia by improving the biological functions of trophoblast via NF-κB pathway

Shuxian Li et al.

Summary: The expression of FPR2 is higher in placental tissues of patients with PE. Knockdown or inhibition of FPR2 can reduce the release of proinflammatory cytokines and reverse the effects of LPS on growth, migration, invasion, angiogenesis, and apoptosis in HTR8/SVneo cells. Activation of the NF-κB signaling pathway is observed in both placental tissues of patients with PE and LPS-treated HTR8/SVneo cells, but this activation is attenuated when FPR2 is knocked down or inhibited.

Summary: This study found that GADD45A plays an important role in coronary artery ischemia/reperfusion injury and has a protective effect on cardiac microvascular endothelial cells. Silencing GADD45A can alleviate apoptosis and functional injury of CMECs by regulating the JNK/p38 MAPK and STAT3/VEGF pathways.

Mitochondrial ROS-mediated ribosome stalling and GCN2 activation are partially involved in 1-nitropyrene-induced steroidogenic inhibition in testes

Jian Li et al.

Summary: In this research, it was discovered that acute exposure to 1-nitropyrene (1-NP) from polluted atmosphere resulted in a decline in testosterone (T) level in men. The study found that the reduction of T contents was caused by the downregulation of steroidogenic proteins in mouse testes and Leydig cells. The mechanism involved the activation of GCN2 and mitochondrial ROS-mediated ribosome stalling, leading to the inhibition of steroidogenic activity.

Ferroptosis: molecular mechanisms and health implications

Daolin Tang et al.

Summary: Cell death can be executed through different subroutines, such as ferroptosis which is an iron-dependent form of non-apoptotic cell death. Ferroptosis can occur through two major pathways, driven by redox imbalance and abnormal expression of redox-active enzymes. The process is precisely regulated at multiple levels, with the transcription factor NFE2L2 playing a central role in anti-ferroptotic defense.

RNA-DNA hybrids regulate meiotic recombination

Xuan Yang et al.

Summary: RNA-DNA hybrids formed at ssDNA ends of DSBs actively regulate meiotic recombination in budding yeast, affecting the efficiency of sporulation and spore viability. The accumulation of RNA-DNA hybrids competes with Rad51/Dmc1, impairs homolog bias, and decreases crossover and noncrossover recombination.

The roles of free iron, heme, haemoglobin, and the scavenger proteins haemopexin and alpha-1-microglobulin in preeclampsia and fetal growth restriction

Lena Erlandsson et al.

Summary: In pregnancies complicated by preeclampsia (PE) and fetal growth restriction (FGR), iron homeostasis is altered, with iron levels significantly correlating with the severity of the diseases. The normal increase in plasma volume during pregnancy is less in PE and FGR, which has a different impact on factors like iron concentration compared to normal pregnancies. Excess iron may promote ferroptosis, contributing to trophoblast stress and lipotoxicity.
