Molecular targeted therapy: A new avenue in glioblastoma treatment

Oula El Atat et al.

Summary: Glioblastoma, or GBM, is a fast-growing and aggressive type of brain tumor that is the most prevalent in adults. Despite advancements in diagnosis and treatment, survival rates have not improved significantly in the past three decades. Genomic signature, radiation exposure, and environmental factors contribute to the development of this tumor.

Christina I. Tsien et al.

Summary: This study evaluated whether reirradiation (re-RT) and concurrent bevacizumab (BEV) improve overall survival (OS) and/or progression-free survival (PFS) compared to BEV alone in recurrent glioblastoma (GBM). The results showed that the combination of re-RT and BEV significantly improved PFS, but had no difference in OS.

Salvage Low-Dose-Rate Prostate Brachytherapy: Clinical Outcomes of a Phase 2 Trial for Local Recurrence after External Beam Radiation Therapy (NRG Oncology/RTOG 0526)

Juanita Crook et al.

Summary: This is the first prospective multicenter trial reporting outcomes of salvage low-dose-rate prostate brachytherapy for local failure after external beam radiation therapy. The 5-year freedom from biochemical failure rate is 68%, comparable to other salvage modalities. Although further local failure is rare (5%), biochemical failure rate climbs to 46% by 10 years.

Multiple direction needle-path planning and inverse dose optimization for robotic low-dose rate brachytherapy

Summary: This article introduces a treatment planning system (TPS) that combines multiple direction needle-path planning with inverse dose optimization algorithms for assisting low-dose rate brachytherapy. The experimental results show that the TPS can generate treatment plans with acceptable dose coverage in a reasonable amount of time, and the results are in good accordance with a commercial TPS.

The Definition and Delineation of the Target Area of Radiotherapy Based on the Recurrence Pattern of Glioblastoma After Temozolomide Chemoradiotherapy

Lin Zheng et al.

Summary: This study examined the recurrence patterns of GBM patients with target delineations that did not intentionally include the T2/FLAIR hyperintensity area outside of the GTV. It was found that excluding the adjacent T2/FLAIR hyperintensity area reduced brain exposure to high-dose radiation without increasing the risk of marginal recurrence. Further clinical trials are warranted to investigate the feasibility of intentionally excluding the T2/FLAIR hyperintensity region outside of the GTV.
