Circadian rhythm-related genes index: A predictor for HNSCC prognosis, immunotherapy efficacy, and chemosensitivity

Summary: This study aimed to establish reliable markers based on biologic clock genes to assess the immunotherapy response and prognosis in HNSCC patients. Through analysis of 502 HNSCC samples and 44 normal samples, 6 circadian rhythm-related genes (CRRGs) strongly associated with HNSCC prognosis were identified and a predictive model was constructed. Patients can evaluate their prognosis and immunotherapy outcome based on the risk score generated by this model.

Summary: The cancer-associated fibroblasts (CAFs) and thrombospondin 2 (THBS2) in the microenvironment of pancreatic ductal adenocarcinoma (PDAC) play important roles in tumor development and prognosis. It has been found that elevated THBS2 expression in PDAC is mainly located in the stroma and is associated with tumor progression and poor prognosis. The study also revealed that cancer cell-secreted TGF-beta 1 activates CAFs to induce THBS2 expression, which then promotes tumor growth and adhesion in PDAC cells.

Profile of circulating extracellular vesicles microRNA correlates with the disease activity in granulomatosis with polyangiitis

Summary: This study characterized the profile of microRNAs trafficked by extracellular vesicles (EVs) in patients with granulomatosis with polyangiitis (GPA). Four differentially expressed microRNAs were identified in EVs from GPA patients, with miR-223-3p being the best discriminator between GPA patients and healthy controls. MiR-664a-3p was able to discriminate between active and remission phases of GPA. The correct classification rate based on multivariate discriminant analysis was 92% for acute phase and 85% for all study participants. Pathway analysis revealed potential target genes of these microRNAs, involved in focal adhesion, mTOR signaling, and neutrophil extracellular traps formation. Two microRNAs positively correlated with disease activity were found to be involved in neutrophil extracellular traps formation and apoptosis inhibition.
