Inflammation in multiple sclerosis: consequences for remyelination and disease progression

IL-33 and HMGB1 modulate the progression of EAE via oppositely regulating each other

Summary: The dynamic expression and release of IL-33 and HMGB1 were examined in different stages of EAE in vivo, revealing their interaction in EAE progression. HMGB1 dominated the pre-onset stage, while IL-33 dominated the peak stage of EAE. Blocking extracellular HMGB1 and conditional knockout of HMGB1 in astrocytes reduced IL-33 release, indicating a promotion of IL-33 release by HMGB1. Further, IL-33 reduced HMGB1 release from primary astrocytes in vitro. Overall, IL-33 and HMGB1 in the CNS jointly participate in EAE progression, with IL-33 potentially contributing to the self-limiting of EAE by inhibiting HMGB1.
