Treatment with quercetin inhibits SARS-CoV-2 N protein-induced acute kidney injury by blocking Smad3-dependent G1 cell-cycle arrest

Wenjing Wu et al.

Summary: Increasing evidence suggests that SARS-CoV-2 can cause acute kidney injury (AKI) in COVID-19 patients, but the mechanisms are unclear and treatments are ineffective. This study found that overexpression of the SARS-CoV-2 N gene in the kidneys of diabetic mice led to AKI, and treatment with quercetin, a traditional Chinese medicine compound, inhibited AKI by blocking the interaction between the SARS-CoV-2 N protein and Smad3. This study highlights the pathogenic role of the SARS-CoV-2 N protein in AKI and identifies quercetin as a potential treatment for COVID-19-induced AKI.

Matrine-induced nephrotoxicity via GSK-3β/nrf2-mediated mitochondria-dependent apoptosis

Tianyang Wang et al.

Summary: This study investigated the roles of oxidative stress and mitochondria in Matrine-induced kidney toxicity. The results showed that Matrine caused nephrotoxicity accompanied by an increase in reactive oxygen species accumulation and mitochondrial dysfunction. Additionally, GSK-3 beta and Nrf2 might serve as promising nephroprotective targets for Matrine-induced kidney injury.

Nickel induces hepatotoxicity by mitochondrial biogenesis, mitochondrial dynamics, and mitophagy dysfunction

Hongrui Guo et al.

Summary: Nickel (Ni) is a hazardous chemical waste that can cause multi-organs toxicity. The liver is the major target organ for Ni accumulation and toxicity, but the exact mechanism is still unclear. This study found that NiCl2 treatment induced hepatic histopathological changes and mitochondrial damage in mice. NiCl2 inhibited mitochondrial biogenesis and disrupted mitochondrial dynamics, leading to increased mitophagy in the liver. The molecular mechanism of NiCl2-induced hepatotoxicity involves dysfunction of mitochondrial biogenesis, dynamics, and mitophagy.

Mitochondria in health, disease, and aging

John S. S. Harrington et al.

Summary: Mitochondria, besides generating ATP through oxidative phosphorylation, also play integral roles in synthesizing metabolic precursors, regulating calcium, producing reactive oxygen species, mediating immune signaling, and inducing apoptosis. Given the wide range of responsibilities, mitochondria are essential for cellular metabolism and homeostasis. Translational medicine has recognized the significance of mitochondrial dysfunction as a predictor of disease, thereby investigating the association between mitochondrial dysfunction and disease pathogenesis. These findings suggest that targeting mitochondria-dependent pathways could be a promising approach for therapeutic interventions in human diseases.

Astilbin targeted Sirt1 to inhibit acetylation of Nrf2 to alleviate grass carp hepatocyte apoptosis caused by PCB126-induced mitochondrial kinetic and metabolism dysfunctions

Yu Xia et al.

Summary: The study found that exposure to PCB126 led to various abnormalities in L8824 cells, including mitochondrial dynamics imbalance, energy metabolism disorder, and promotion of apoptosis. However, treatment with Astilbin reversed these abnormalities and promoted the deacetylation of Nrf2 by interacting with Sirt1, thereby alleviating PCB126-induced apoptosis.

Exploring the Role of NCX1 and NCX3 in an In Vitro Model of Metabolism Impairment: Potential Neuroprotective Targets for Alzheimer's Disease

Alessandra Preziuso et al.

Summary: Alzheimer's disease (AD) is a progressive neurodegenerative disorder that affects elderly people and is characterized by brain cell deterioration. The study found that silencing the NCX3 protein can improve cell viability, increase energy production, reduce oxidative stress damage, and decrease abnormal protein levels associated with AD. This discovery provides a potential strategy for preventing the development of AD.

Decabromodiphenyl ether initiates mitochondria-dependent apoptosis by disrupting calcium homeostasis in mice livers

Siyan Che et al.

Summary: This study investigated the effects of BDE-209 on mouse liver and revealed that it leads to an imbalance of redox and promotes apoptosis with a mitochondria-dependent manner. BDE-209 induced changes in mitochondrial morphology and increased ER-mitochondrial contact. ER stress was involved in the apoptosis process, and there was overloaded mitochondrial Ca2+ in hepatocytes of BDE-209 treated mice.

Quercetin alleviates acute kidney injury by inhibiting ferroptosis

Yue Wang et al.

Summary: The study demonstrated that quercetin (QCT), a natural flavonoid, inhibited ferroptosis in renal proximal tubular epithelial cells by reducing lipid ROS levels and increasing glutathione (GSH) levels. QCT also ameliorated acute kidney injury induced by ischemia-reperfusion (I/R) or folic acid (FA). Furthermore, RNA-sequence analysis revealed ATF3 as a key factor in QCT's inhibition of ferroptosis. Additionally, QCT inhibited the recruitment of pro-inflammatory macrophages in AKI.
