5-aminolevulinic acid-loaded dissolving microneedle array for photodynamic therapy of rheumatoid arthritis on rats

Summary: Photodynamic therapy (PDT) is a noninvasive technique that can be used to treat rheumatoid arthritis (RA) by delivering photosensitizers using a microneedle array. The therapy effectively inhibits the migration ability and induces apoptosis of RA fibroblast-like synoviocytes. Additionally, in a rat model of adjuvant arthritis, PDT using 5-aminolevulinic acid-loaded dissolving microneedle array demonstrates significant therapeutic effects by regulating cytokine expression. It suggests that 5-ALA@DMNA-mediated PDT may be a potential therapy for RA.

Dissolving microneedle patch-assisted transdermal delivery of methotrexate improve the therapeutic efficacy of rheumatoid arthritis

Summary: This study developed a dissolving microneedle patch (DMNP) for transdermal delivery of methotrexate (MTX) to treat rheumatoid arthritis (RA). The DMNP demonstrated excellent morphology and mechanical strength, and significantly increased the transdermal permeation of MTX compared to cream. In vivo experiments showed that MTX-loaded DMNP effectively alleviated RA symptoms and inhibited inflammatory response. This transdermal drug delivery system has the potential to improve the safety, convenience, and efficacy of MTX treatment for RA.

This article discusses factors that contribute to the infection risk and emphasizes the importance of dialogue between stakeholders for the development of safe and effective MAP products.

Synthetic biology-instructed transdermal microneedle patch for traceable photodynamic therapy

The patch is loaded with tumor acidity-responsive nanoparticles containing 5-aminolevulinic acid and catalase, which can reverse tumor hypoxia and enhance the accumulation of protoporphyrin IX. The patch also allows for fluorescence/photoacoustic duplex imaging to monitor intratumoral oxygen saturation and protoporphyrin IX metabolic kinetics.
