Alternative oxidase 2 influences Arabidopsis seed germination under salt stress by modulating ABA signalling and ROS homeostasis

Polymorphisms in alternative oxidase genes from ecotypes of Arabidopsis and rice revealed an environment-induced linkage to altitude and rainfall

Summary: We investigated the SNP in alternative oxidase (AOX) genes and their association with ecotype origins. We detected non-synonymous SNPs in the AOX ORF, leading to amino acid changes in AOX1c, AOX1d, and AOX2 in Arabidopsis, and AOX1c in rice. The AA changes in Arabidopsis were linked to high rainfall and altitude, while all changes in Arabidopsis and rice AOX were associated with high altitude and rainfall. Comparative 3D modeling revealed structural differences between mutant AOX and their respective references. Molecular docking analysis showed better enzyme-substrate binding affinities for the identified structures. Therefore, the majority of AA changes found in the ecotypes indicate environment-related, more efficient AOX activity.

Furthermore, we found that OsHIPL1 interacts with the aquaporin OsPIP1;1, affecting water uptake and promoting rice seed germination.

Alternative oxidase (AOX) 1a and 1d limit proline-induced oxidative stress and aid salinity recovery in Arabidopsis
