Study of tumor necrosis factor receptor in the inflammatory bowel disease

Summary: Ulcerative colitis (UC) and Crohn's disease (CD) are types of Inflammatory Bowel Diseases (IBD) characterized by bowel necrosis and the involvement of enteric neurons and glial cells. The main inflammatory mediator is tumor necrosis factor-alpha (TNF-alpha), which is also associated with the production of other cytokines like interleukins (IL). TNF-alpha acts through two receptors, TNFR1 and TNFR2, which have been detected in IBD and their expression correlates with disease activity. Biologic agents targeting TNF-alpha have revolutionized IBD treatment by inducing and maintaining disease remission in a majority of patients.

Anti-inflammatory effect of metformin against an experimental model of LPS-induced cytokine storm

Summary: Cytokine storm is a major cause of death in COVID-19 patients, and metformin has been shown to inhibit proinflammatory cytokines. This study examined the anti-inflammatory effect of oral and intraperitoneal metformin in a mouse model of LPS-induced cytokine storm. Both routes of metformin administration significantly reduced IL-1, IL-6, and TNF-alpha levels, with intraperitoneal metformin being more effective in reducing IL-6 levels. Metformin shows potential in ameliorating diseases caused by elevated cytokine levels.

The adult gut microbiota is regulated by diet and host factors, with oxygen and nitrate availability shaping the bacterial community in different parts of the gut. Poor diet weakens host control mechanisms, potentially leading to dysbiosis. Therefore, quantifying host parameters that control microbial growth could offer alternative approaches to define and remediate dysbiosis.
