Targeting PFKL with penfluridol inhibits glycolysis and suppresses esophageal cancer tumorigenesis in an AMPK/FOXO3a/BIM-dependent manner

Cancan Zheng et al.

Summary: The study shows that penfluridol inhibits glycolysis and induces apoptosis in esophageal squamous cell carcinoma (ESCC). It targets AMPK and phosphofructokinase, liver type (PFKL), and high PFKL expression is associated with poor prognosis. Penfluridol may be a potential therapeutic option for ESCC.

MicroRNA-342-3p is a potent tumour suppressor in hepatocellular carcinoma

Ronja-Melinda Komoll et al.

Summary: This study identified miR-342-3p as a tumor suppressor miRNA in HCC, showing its therapeutic efficacy in attenuating tumor development and prolonging survival in different HCC mouse models. The downregulation of miR-342-3p in HCC tumors from patients compared with non-tumor tissues, inversely correlating with MCT1 expression, was also observed.

Identification of lysine isobutyrylation as a new histone modification mark

Zhesi Zhu et al.

Summary: Lysine isobutyrylation (Kibu) is a newly discovered posttranslational chemical modification on nuclear histones, derived from isobutyryl-CoA. Various histone acetyltransferases exhibit lysine isobutyryltransferase activity in vitro, with p300 being a major regulator of histone isobutyrylation levels in cells. The X-ray crystal structures of HAT1 in complex with isobutyryl-CoA provide insight into HAT-catalyzed isobutyrylation, and RNA-Seq profiling shows that isobutyrate greatly affects gene expression in pivotal biological pathways.

Autophagy, an accomplice or antagonist of drug resistance in HCC?

Yafei Wu et al.

Summary: Hepatocellular carcinoma, a highly lethal malignancy, shows poor response to drug treatment with frequent drug resistance. Autophagy plays a crucial role in HCC drug resistance, interacting with cancer stem cells, tumor-derived exosomes, and noncoding RNA. Clinical trials focusing on autophagy inhibition combined with anticancer drugs are of interest.

Protein Lipidation by Palmitoylation and Myristoylation in Cancer

Chee Wai Fhu et al.

Summary: Protein lipidation, a posttranslational modification involving lipid moieties, plays a crucial role in modulating the properties and interactions of proteins, closely linked to cellular metabolism. The two most common protein lipid modifications, palmitoylation and myristoylation, have significant implications in regulating cancer metabolism and progression. Dysregulation of protein lipidation is strongly associated with various diseases, highlighting the importance of understanding its roles in health and disease.

Summary: Azelastine, an anti-allergic drug, was found to significantly inhibit CRC cell proliferation but not in ARF1-deficient cells. High ARF1 expression was associated with advanced stages and poor survival of CRC, promoting colon tumorigenesis through interaction with IQGAP1.
