The flip-flop configuration of the PABP-dimer leads to switching of the translation function

Sohyun Gu et al.

Summary: PABP is a translation initiation factor that interacts with the poly(A) tail of mRNAs, stimulating translation by interacting with eIF4G. It is present in higher quantities than mRNA and eIF4G in cells. Research showed that RNA-free PABPs dimerize in a head-to-head configuration, interfering with PABP-eIF4G interaction, and Poly(A) RNA can convert the PABP-PABP complex into a Poly(A)-PABP complex, facilitating PABP-eIF4G interaction.

Summary: This study reports single-molecule methods to visualize each step of miRNA-mediated gene silencing inside cells. It is found that miRNAs preferentially bind to translated mRNAs and induce translational repression and mRNA decay after binding to mRNAs.

Summary: Eukaryotic translation is a complex process involving various translation factors. A study found that the co-localization of eIF4E, eIF4G, and PABP proteins on mRNA is minimal, suggesting that the communication between them during translation may be transient.

mRNA- and factor-driven dynamic variability controls eIF4F-cap recognition for translation initiation

Burak Cetin et al.

Summary: Studies have shown that the interaction between eIF4F and mRNA is regulated by mRNA length, cap-proximal secondary structure, eIF4A, and ATP. The activities of eIF4G and eIF4A accelerate the association rate of eIF4E with mRNA, playing a crucial role in translation.

Imaging translational control by Argonaute with single-molecule resolution in live cells

Charlotte A. Cialek et al.

Summary: The authors developed live-cell biosensors to investigate the impact of Argonaute proteins on single-mRNA translation dynamics. They found that Argonaute association gradually silences translation and leads to the clustering and coalescence of mRNA with P-bodies, maintaining a translationally silent state.

Summary: This study combines live-cell spectroscopy and single-particle tracking to investigate the binding dynamics of mRNA translation initiation factors. By quantifying their diffusion characteristics, the assembly and disassembly sequence of initiation factors and the clustering of translation in neurons are revealed.

Summary: This study demonstrates a new method to visualize endogenous genomic loci using synthetic zinc-finger proteins and frankenbodies for signal amplification, enabling the simultaneous observation of the dynamics of two loci in single living cells. The results show a close association between the two loci and their separation upon the triggered degradation of the cohesin subunit RAD21.
