A novel fluorescent probe with ACQ-AIE conversion by alkyl chain engineering for simultaneous visualization of lipid droplets and lysosomes

Summary: A series of novel AIE probes were successfully designed and synthesized in this study, and the transformation from ACQ to AIE effect was achieved through the alkylation of 4-aminonaphthalimide. Among them, the AIE probe NR-Lyso-IV exhibited excellent emission properties and solubility, and it was able to simultaneously visualize lipid droplets and lysosomes in HeLa cells and zebrafish.

N-Cyanorhodamines: cell-permeant, photostable and bathochromically shifted analogues of fluoresceins

Red AIE Luminogens with Tunable Organelle Specific Anchoring for Live Cell Dynamic Super Resolution Imaging

Summary: This study presents two efficient red-emitting aggregation-induced emission (AIE) luminogens with off-on fluorescence characteristic for specific lysosomes and mitochondria targeting. The AIE luminogens show good biocompatibility, excellent photostability, and a Stokes' shift larger than 100 nm, enabling high fidelity imaging of lysosomes and mitochondria. Moreover, dynamic lysosomal fusion and mitochondrial fission are imaged with a high resolution of 65.6 nm using stimulated emission depletion (STED) imaging. Additionally, interactions between lysosomes and mitochondria during mitophagy are recorded, providing practical guidance for designing specific organelle targeting probes for live cell dynamic super-resolution imaging.
