Hypoiodite-Catalyzed Oxidative Umpolung of Indoles forEnantioselective Dearomatization

Summary: In this study, we reported the oxidative umpolung of 2,3-disubstituted indoles towards enantioselective dearomative aza-spirocyclization using chiral quaternary ammonium hypoiodite catalysis. Mechanistic studies revealed the reactivity of C3 of indoles by iodination of the indole nitrogen atom. Furthermore, the introduction of pyrazole as an electron-withdrawing auxiliary group at C2 suppressed a competitive dissociative racemic pathway, enabling enantioselective spirocyclization to proceed and providing access to both spiropyrrolidines and four-membered spiroazetidines that are otherwise difficult to obtain.

A ring expansion strategy towards diverse azaheterocycles

Summary: This study presents a novel ring expansion strategy involving the cross-dimerization of different strained rings through synergistic bimetallic catalysis, providing an efficient and scalable method for assembling diverse N-heterocycles. Preliminary mechanistic studies reveal that the C-C bond of strained ring ketones is cleaved by the Pd-0 species during the reaction.

Rapid synthesis of azepinoindole derivatives from tryptamine sulfonamides and bromoallyl sulfones via an acid-mediated cyclization and rearrangement

Summary: The novel acid-promoted cyclization-rearrangement method was used to synthesize the azepinoindole framework from tryptamine-derived N-sulfonyl enamines, which were prepared through a cesium carbonate mediated vinylic substitution reaction. The steric bulk of the N-sulfonyl group on tryptamine was found to be crucial in determining the reaction outcome.
